摘  要:目的探讨Noggin基因修饰的神经干细胞移植到大鼠局灶性脑缺血处的迁移、分化及对神经功能损伤的修复作用。方法选取SD大鼠50只建立大脑中动脉梗塞模型,随机分为对照组10只,神经干细胞移植组、Noggin-神经干细胞移植组各20只,分别移植入10μl PBS、BrdU标记神经干细胞及Noggin-神经干细胞。移植前及移植后7d、14d、21d对各组大鼠进行NSS评分。25d时处死各组大鼠,脑组织免疫组化染色观察植入细胞的存活及分布情况。结果移植后各组NSS评分均下降,Noggin-神经干细胞移植组评分第14d、21d显著低于对照组及神经干细胞移植组,差异具有统计学意义(P<0.05)。Noggin-神经干细胞移植组BrdU阳性细胞(126.8±5.46)个/HP,多于神经干细胞移植组(85.5±4.7)个/HP,差异具有统计学意义(P<0.05)。结论 Noggin基因能促进神经干细胞在缺血区域的增殖、迁移与分化,对神经功能损伤的修复具有积极作用。Objective To investigate the effects of transplanting Noggin gene modified neural stem cell migration in-to rats with focalcerebral ischemia.Methods Established middle cerebral artery occlusion model of 50 rats.Randomly divided them into control group of 10,neural stem cells transplantation group,Noggin-neural stem transplantation cell group with 20 in each group.Transplanted into 10 μL PBS,BrdU labeled neural stem cells and neural stem cells of Nog-gin.Before and on the 7,21,14 d after the transplantation,record NSS score of the rats.rats were killed on the 25 d, and observe the survival and distribution of transplanted cells.Results NSS score were decreased fter transplantation. The 14 d,21 d scores were significantly lower than control group and neural stem cell transplantation group.Noggin-neural stem cell transplantation group had BrdU positive cell (126.8±5.46)/HP,more than neural stem cells trans-plantation group (85.5±4.7)/HP,all with significant difference (P 〈0.05).Conclusion Noggin gene can promote the proliferation,migration and differentiation of neural stem cells in the ischemic region,having a positive effect on neurological functional recovery.
