PD-1及PD-L1在脓毒症小鼠外周血中性粒细胞的表达

摘　　要：目的探讨脓毒症小鼠外周血中性粒细胞膜分子PD-1及配体PD-L1表达。方法 45只C57小鼠随机分为6 h,24h模型组及假手术组。6 h模型组10只,24 h模型组15只。假手术组共20只,6 h、24 h时间点各10只。盲肠结扎穿孔法造模,造模成功后分别于6 h、24 h取外周血,采用流式细胞术检测各组不同时间点的中性粒细胞PD-1及PD-L1表达。结果6 h组的PD-1及PD-L1表达较假手术组未见升高(P>0.05),24 h组PD-L1表达较假手术组明显升高(P<0.01)、较6 h组表达明显升高(P<0.01)。24 h组PD-1表达较6 h组和假手术组无升高(P>0.05)。结论小鼠脓毒症早期中性粒细胞PD-L1表达增高,PD-1表达明显无变化。Objective To explore the PD-1 and PD-L1 ligand expression on peripheral blood neutrophils in septic mice. Methods Forty-five C57 mice were divided randomly into 6 h model group（n = 10）, 24 h model group（n = 15） and sham group（n =20）. The mice in sham group were sacrificed at 6 h and 24 h, respectively. Sepsis model was induced by cecal ligation and puncture. Peripheral blood was obtained at 6 h and 24 h to detect PD-1 and PD-L1 expression on neutrophils by flow cytometry. Results Expression of PD-1 and PD-L1 in 6 h model group were not significantly increased compared with sham group（ P 〉 0.05 ）. PD-L1 expression in 24 h model group was significantly higher than that in sham group （ P 〈 0. O1 ） and 6 h model group （ P 〈 0.01 ）. PD-1 expression in 24 h model group was not higher than that in sham group （ P 〉 0.05 ） and 6 h model group（ P 〉 0.05 ）. Conclusion PD-L1 expression on neutrophils is increased,but PD-1 shows no significant change in early septic mice.
