摘  要:目的讨论stil基因表达上调对HEK293T细胞周期的影响。方法构建过表达STIL质粒,转染HEK293T细胞,PI/RNase双染色后流式细胞仪检测细胞周期变化。结果空白对照组与转染对照组相比,stil基因表达上调导致HEK293T细胞各个周期细胞比例无显著差异(P>0.05);实验组与转染对照组相比,在过表达24h、48h、72h后,G1期细胞比例显著下降,S期细胞比例明显增加(P<0.05)。结论 stil基因表达上调影响了HEK293T细胞周期的正常进程,导致细胞周期紊乱。Objective:Discuss the effect of HEK293 T cell cycle caused by the up-regulation of stil gene expression. Method:After over-expression plasmids transfect HEK293 T cell,stil will be PI/RNase double stained at three time points respectively. Then the changes of cell cycle will be detected by flow cytometry. Result:Blank control group and transfection control group,the number of HEK293 T cells in each cycle has no significant difference(P0.05). Experimental group compared to transfection control group,after the over-expression of 24 h,48h,72 h,the cell proportion in G1 phase declined significantly,but the cell proportion in S phase increased significantly(P0.05). Conclusion:The up-regulation of stil gene expression affects the normal process of HEK293 T cell cycle and causes the disorder of cell cycle.
