化疗药物顺铂和阿霉素联合应用体外研究

摘　　要：目的:定量分析抗肿瘤药物顺铂和阿霉素联合应用的协同、相加或拮抗作用,筛选顺铂和阿霉素联合应用的最佳配比。方法:不同配比的化疗药物顺铂和阿霉素处理HepG2细胞24h,MTT法测定HepG2细胞的增殖抑制作用,Cbou-Talalay法计算药物的联合指数和两种药物对HepG2细胞的敏感性,分析药物不同配比的相互作用,筛选顺铂和阿霉素联合应用的最佳配比。结果:顺铂、阿霉素单独和联合应用对HepG2细胞的抑制率呈剂量依赖性量效关系;两种药物以小差别比例配伍时,小剂量联合应用呈现拮抗作用,大剂量联合应用呈协同作用;两种药物以大差别比例配伍时,联合效应结果相反;综合药物联合指数和两种药物对HepG2细胞的敏感性,优选出顺铂和阿霉素联合应用的最佳配比为1∶15。结论:化疗药物顺铂和阿霉素不同配比联合应用均可产生增强互补、减毒增效的作用。Objective：Quantitative evaluation of the relationship between anticancer drugs cisplatin and doxorubicin combined effect of screening the best ratio of combination therapy.Methods：Different ratios of cisplatin and gemcitabine＇s inhibition rates of HepG2 cells was measured by MTT assay.Utilize median-effect principle to calculate combination index and the sensitivity of the two drugs on HepG2 cells,and evaluate drug interactions.Results：Cisplatin and Adriamycin＇s inhibition rate of HepG2 cells in a dose-dependent relationship between dose no matter alone and in combination.Small doses of the drug combination,showing a synergistic effect,and highdose combination is showing antagonism.Integrated drug combination index and the sensitivity of the two drugs on HepG2 cells,the best ratio optimized cisplatin and adriamycin is 1：15.Conclusion：Median-effect principle is often used to evaluate drug interactions in vitro,which evaluation can be combined with the drug,the best ratio of anticancer drugs combined screening applications.
