摘 要:目的研究超声介入法建立小鼠肝细胞癌模型的可行性及稳定性,并探讨超声在肝细胞癌模型中的应用价值。方法在超声引导下将细胞浓度为2×107/ml的H22小鼠肝癌细胞悬液注入30只BALB/C小鼠肝左叶,于注射后10 d和20 d行肝脏超声检查,并在20 d超声检查后行剖腹探查,作HE染色,观察成瘤率、肿块的大小及超声表现。结果超声介入法接种20 d后,超声检查可见29只小鼠肝内肿瘤生成,可探及瘤周血流,瘤块最大径平均(8.4±0.3)mm,与剖腹探查结果(8.6±0.2)mm比较,差异无统计学意义。肝细胞癌模型成功率为96.7%,病理证实均为肝细胞癌。结论超声介入法建立小鼠肝细胞癌模型方法简便、微创、成功率高,可为肝细胞癌的研究提供理想模型。Objective To study the feasibility and stability of hepatocellular carcinoma model established in mouse with the guidance of ultrasound, and to explore the value of ultrasonography in hepatocellular carcinoma model. Methods The type of H22 cells, whose concentration was 2×10^7/ml, were injected into the left liver of 30 mice with the guidance of ultrasound. After injection, the mice were examined by ultrasound on the tenth and twentieth day, and implemented laparotomy on the twentieth day. The tumors were examined pathologically. The formation rate and growth of tumors were evaluated, and the ultrasonic features of tumors were observed. Results The formation rate of implanted tumors was 96.7%(29/30), and the peritumoral blood fl ow could be detected. The diameters measured by ultrasound and explorative laparotomy were(8.4±0.3) mm and(8.6±0.2) mm, respectively(P 〉0.05). The pathological examination showed that the tumors were hepatocellular carcinoma. Conclusion Establishing mouse model of hepatocellular carcinoma by ultrasound-guided is simple and minimally invasive, and it provides an ideal model of high formation rate for the study of hepatocellular carcinoma.
