摘　　要：目的探讨表皮生长因子受体(epidermal growth factor receptor,EGFR)在骨肉瘤中的蛋白表达及基因扩增,并评估其在骨肉瘤发生、发展和预后中的价值。方法以石蜡包埋组织芯片为材料,分别采用免疫组织化学PV-9000两步法与荧光原位杂交(fluorescent in situ hybridization,FISH)技术检测32例骨肉瘤及10例骨软骨瘤中EGFR蛋白表达及基因扩增。结果 EGFR蛋白在骨肉瘤、骨软骨瘤中的表达阳性率分别为84.4%、20.0%;扩增阳性率分别为75%、20%;骨肉瘤中EGFR蛋白表达、基因扩增阳性率与骨软骨瘤相比较,差异具有统计学意义(P<0.05);在骨肉瘤中EGFR蛋白表达、基因扩增阳性率与患者的年龄、性别、肿瘤部位无相关性(P>0.05),与肿瘤转移、临床分期相关(P<0.05)。结论 EGFR与骨肉瘤的发生、发展有关,可以将其作为判断骨肉瘤生物学行为的重要参考指标。Objective To investigate the expression of EGFR protein and gene amplification in osteosarcoma and evalua-tion of EGFR in osteosarcoma occurrence,development and prognosis. Methods PV-9000 two step method and in situ chemi-cal hybridization（FISH）were used to detect the expression of EGFR protein,gene amplification in 32 cases of osteosarcoma and 10 cases of osteochondroma in paraffin-embedded tissue chip material. Results The positive rate of EGFR expresion in osteosarcoma,osteochondroma were 84. 4% 、20. 0% . The positive rate of EGFR gene amplification were 75% and 20% . There were significant differences between osteosarcoma and osteochondroma in the expression and amplification of EGFR. The ex-pression and amplification of EGFR in osteosarcoma had no correlation with age ,gender ,or the location of the tumor（ P 〉0. 05）,but were correlated with metastasis and clinical stage（P〈0. 05）. Conclusion EGFR is closely related to the occur-rence and development of osteosarcoma,and can be considered as an important index for determing the biological behavior of osteosarcoma.
