MIMECAN基因、细胞核因子κB和白细胞介素24与子宫内膜癌的相关性研究

摘　　要：目的探讨MIMECAN基因、细胞核因子(NF)-κB和白细胞介素(IL)-24在子宫内膜癌中的表达及其关系。方法选择2008年1月至2013年1月于徐州医学院附属淮安医院妇产科住院治疗的120例子宫内膜癌患者为研究对象,将其纳入子宫内膜癌组(n=120),再按照国际妇产科联盟(FIGO)病理学分期标准将其进一步分为4个亚组,分别为Ⅰ期亚组(n=45)、Ⅱ期亚组(n=30)、Ⅲ期亚组(n=24例)和Ⅳ期亚组(n=21)。选择同期在本院行健康体检的40例正常妇女为对照组(n=40)。采用酶联免疫吸附测定法(ELISA)检测各组血清MIMECAN基因、NF-κB和IL-24水平。本研究遵循的程序符合徐州医学院附属淮安医院妇产科人体试验委员会所制定的伦理学标准,得到该委员会批准,分组征得受试对象本人的知情同意,并与之签署临床研究知情同意书。结果 1子宫内膜癌组各亚组患者血清MIMECAN基因、NF-κB和IL-24水平均明显高于对照组,差异均有统计学意义(P<0.05),且各亚组间两两比较,差异也有统计学意义(P<0.05)。2MIMECAN基因、NF-κB和IL-24与子宫内膜癌严重程度呈正相关关系(r=0.69,0.73,0.82;P<0.05)。3子宫内膜癌组MIMECAN基因和NF-κB呈正相关关系(r=0.76,P<0.05);NF-κB和IL-24也呈正相关关系(r=0.78,P<0.05)。结论 MIMECAN基因、NF-κB和IL-24相互作用可能促进子宫内膜癌的发生、发展。Objective To explore the expression of MIMECAN gene, cell nuclear factor（NF）-κB, interleukin（IL）-24 and their relationship with endometrial carcinoma. Methods From January 2008 to January 2013,a total of 120 hospitalized patients with endometrial carcinoma in Huai＇an Affiliated Hospital of Xuzhou Medical College were included in the study as research group（n= 120）,they were further divided into stage Ⅰ subgroup（n= 45）, stage Ⅱ subgroup （n： 30） ,stage Ⅲsubgroup （n= 24） and stage IV subgroup （n= 21） according to the International Federation of Gynecology and Obstetrics（FIGO） pathological stages. Meanwhile, another 40 healthy women were chosen as control group（n=40）. The expression of MIMECAN gene,NF-κB, IL-24 levels were detected by enzyme-linked immunosorbent assay （ELISA）. The study protocol was approved by the Ethical Review Board of Investigation in Human Being of Affiliated Hospital of Xuzhou Medical College. Informed consent was obtained from each participants. Results （1） The levels of MIMECAN gene, NF-~B, IL-24 in research group were obviously higher than those of control group, and the differences were significant （P〈0. 05）. There also were significant differences among four subgroups in levels of MIMECAN gene,NF-κB, IL-24（P（0. 05）. OThere were positive correlation between levels of MIMECAN gene, NF-κB,IL-24 and the severity of endometrial carcinoma（r=0.69, 0. 73,0. 82;P〈0. 05）. ③The NF-κB was positively correlated with MIMECAN gene and IL-24 in research group（r=0.76,0.78; P〈0.05）. Conclusions MIMECAN gene, NF-κB, IL-24 interaction might promote the development of endometrial carcinoma.
