摘 要:目的利用454高通量测序技术,分别对高龋和无龋受试者口腔唾液微生物进行测序,并分析其群落结构及其与龋病的相关性。方法分别采集10例高龋患者和10例无龋受试者口腔唾液样本。提取样本细菌总基因组DNA,进行16Sr DNA的PCR扩增,454焦磷酸测序。结果共检出201种细菌,归属于19个细菌门类。两组优势菌群含量差异无显著性(P>0.05)。在不同个体唾液样本中,高龋组不同个体唾液微生物多样性高于无龋组(P<0.05)。共有14种细菌含量在组间差异具有显著性(P<0.05)。结论人唾液微生物群落具有广泛的多样性,不同个体群落结构差异较大,但拥有共同的核心微生物;龋病并非因某些致龋微生物所形成,而是由于群落结构的多样性及其变化。Objective Using 454 pyrosequencing technology,the high caries and caries free subjects with oral salivar-y microorganisms were sequenced,and analyzed the community structure and its correlation with caries. Method Col-lect 10 high caries patients and 10 caries free subjects saliva samples. Extraction of total genomic DNA samples of bacteria,16SrDNA PCR amplification,454 pyrosequencing. Result There were 201 kinds of bacteria,belonging to 19 phyla of bacteria. No statistically significant difference between the two groups of dominant bacteria content ( P〉0. 05). In different individual salivary samples,high diversity in different individual salivary microbial caries group is higher than that of caries free group(P〈0. 05). There are 14 kinds of bacteria content have significant difference between groups(P〈0. 05). Conclusion The broad diversity of human salivary microbial community,individual differences in community structure in different large,but has the core of microbial common;caries formation are not because of some cariogenic microorganisms,but because the community structure diversity and change.
