局灶性脑缺血大鼠凝血酶与炎症因子相关性研究

摘　　要：目的研究大鼠局灶性脑缺血后凝血酶(Thrombin)及其受体(PAR-1)和炎症因子单核细胞趋化蛋白(MCP-1)、核转录因子κB(NF-κB)的时间变化规律过程及其相关性。方法线栓法建立左侧大脑中动脉闭塞大鼠模型(MCAO),分别在大鼠缺血后1h,6 h,24 h,48 h,72 h进行行为学评分,断头取脑,荧光定量实时PCR(Real-timePCR)方法检测脑组织内Thrombin、PAR-1及MCP-1、NF-κB的mRNA表达。结果大鼠缺血后,荧光定量实时PCR分析提示:Thrombin、PAR-1、NF-κB及MCP-1的mRNA在造模后开始明显升高,24 h达到高峰,后逐渐下降。结论脑缺血后,脑组织内炎症因子表达增多,其表达与凝血酶表达在时间上有相关性,凝血酶可能通过诱导脑组织表达和产生炎症因子,促发炎症反应,加重脑细胞的损害。Objective To observe thrombin and its receptor （PAR 1）and inflammation factor monocyte chemotactic protein （MCP 1）, and nuclear factor kappa B（NF kappa B）’s time variation process and their correlation in rats after focal cerebral ischemia.Methods Line plug method to establish the left middle cerebral artery occlusion （MCAO）rat model,respectively in each time point （ischemia of rats after 1 h,6 h,24 h,48 h,72 h）,the behavior score,then beheaded in brain,real time fluorescent quantitative PCR （real time PCR）method to detect brain thrombin,and MCP 1 PAR 1,the NF kappa B mRNA expression.Results After ischemia in rats, Thrombin mRNA,PAR 1 mRNA,NF kappa B mRNA and MCP 1 mRNA began increased significantly after making touch,and reach to peak in 24 h,then gradual y decline.Conclusion After cerebral ischemia,the expression of inflammation factors increased within the brain tissue,So did the expression of thrombin.So thrombin may promote inflammation and damage brain cel s by induce brain tissue express and produce inflammation factors.
