摘 要:目的采用响应面法优化乌梅总有机酸的提取条件,并对其氧化活性进行评价。方法在单因素试验基础上,采用响应面分析法研究了液固比、浸泡时间、提取时间对乌梅总有机酸提取率的影响;并通过乌梅总有机酸提取液对DPPH自由基的清除作用来研究其抗氧化活性。结果乌梅总有机酸的最佳提取条件:液固比14∶1(m L∶g),浸泡时间70 min,提取时间1.5 h,提取2次。在此条件下,乌梅总有机酸的提取率为35.21%;乌梅总有机酸清除DPPH自由基的半数抑制浓度IC50值为1.788 mg/m L(以生药计)。结论优化所得工艺条件简单可行,可用于乌梅中总有机酸的提取;乌梅总有机酸具有较强的抗氧化作用。Objective To optimize the extraction conditions of total organic acids from Fructus mume and evaluate the antioxidant activity. Methods Based on the single-factor tests,the effects of liquid-solid ratio, soaking time and extraction time on the extraction yield of total organic acids from Fructus mume were studied by response surface methodology. Meanwhile, the antioxidant activity of total organic acids was assessed by scavenging assays of DPPH free radicals. Results The optimal extraction conditions were as follows: extracting with water for twice and 1.5 hours each time, liquid-solid ratio of 14 : 1 ( mL :g) and soaking for 70 min. Under the optimal conditions, the maximum extraction rate of total organic acids was 35. 21%. The IC5o of the total organic acids against DPPH free radicals were 1.788 mg/mL. Conclusion The optimum extraction process was simple and feasible, and it could be used to extract total organic acids from Fructus mume. The total organic acids from Fructus mume showed strong antioxidant capacity.
