摘  要:为观察Toll样受体/核转录因子-κB(TLRs/NF-κB)信号通路在溃疡性结肠炎大鼠结肠中表达的相关性,选取成年健康Wistar大鼠40只,分成实验组30只与对照组10只。对照组正常饲养,实验组根据右旋葡聚糖硫酸钠(DSS)加激怒法建立溃疡性结肠炎模型。造模成功后,采用免疫组化方法观察对比两组结肠组织中TLR-2、TLR-4、NF-κB表达的差异及观察TLR-2、TLR-4和NF_KB的相关性。结果显示,实验组中TLR-2、TLR-4、NF-κB的表达均较对照组中高,且TLR-2、TLR-4、NF-κB的表达与病理分级呈相关性。结果表明,TLRs/NF-κB信号通路异常导致肠黏膜的炎症反应,并与病情严重程度相关。This study was to observe the expression of Toll-like receptors/nuclear transcripation factor-κB (TLRs/NF-κB) signaling pathway in rats with ulcerative colitis,40 healthy adult Wistar rats were selected and divided into experimental group ( n = 30)and control group( n = 10).Control group received normal feeding, experimental group were established model by dextran sulphate sodium(DSS) plus irritating meth- ods.Immunohistochemistry was used to observe the expression of TLR-2, TLR-4 and NF-κB difference, and their correlation in rectal tissue.The results showed that the expression of TLR-2,TLR-4 and NF-κB was relatively high in experimental group than in control group;and there was a correlation between the expression of TLR-2,TLR-4,NF-κB and pathological grade.It is concluded that TLRs/NF-κB signaling pathway abnormal can induce inflammation in intestinal mucosa,which is associated with disease severity.
