摘  要:目的:考查番茄红素(lycopene)对原代培养大鼠皮层神经细胞低糖氧损伤的保护作用,并对其可能的线粒体作用进行分析。方法:原代培养大鼠皮层神经细胞于培养d 7分为正常组、低糖无氧损伤组(95%N2/5%CO2饱和的低糖DMEM培养基)、番茄红素低、中、高剂量组(10-7,10-6和10-5mol·L-1)。番茄红素在低糖氧损伤的同时加入培养液。观察细胞坏死与凋亡、ATP水平、线粒体总量、线粒体膜电位及线粒体呼吸功能。结果:番茄红素能明显减少低糖氧损伤导致的神经细胞坏死与凋亡,提高细胞内ATP水平和线粒体总量,提高线粒体膜电位水平,并明显改善线粒体呼吸功能。结论:番茄红素对原代培养大鼠皮层神经细胞低糖氧损伤具有明显保护作用,这一作用与其保护和改善线粒体呼吸功能和能量代谢密切相关。Objective: To investigate the protective effect of lycopene on hypoxia/hypoglycemie injury in primary culiured cortical neurons, and the possible mitoehondrial actions. Methods: Primary euhured cortical neu- rons were subjected to hypoxia/hypoglycemia(95% N2/5% CO2 saturated DMEM culture medium, low glucose), and lycopene at 10-7, 10-6 and 10-5 mol.l-I was added at the same time. The necrosis and apoptosis of neurons were detecled, and the ATP content, the mitochondrial mass, mitochondrial membrane potential and mitochondrial respiration were measured for the analysis of mitochondrial mechanisms. Results: Lycopene remarkably decreased the neuronal necrosis and apoptosis, and increased the ATP content and the milochondrial mass. The mitoehondrial membrane potential and the mitoehondrial respiration were also found to be improved by the use of lycopene. Con- clusion: l,ycopene has a pronounced protective effect on hypoxia/hypoglycemic injury in primary cultured rat corti- cal neurons, and the mitochondrial action is clearly involved, including the improvement of mitochondrial respira- tion and energy supplement.
