摘 要:目的检测子宫内膜样腺癌组织中LRP16的表达,探讨其与人子宫内膜样腺癌的关联及其与E-cadherin的调控关系。方法 RT-PCR法检测子宫内膜样腺癌组织中LRP16 mRNA的表达;免疫组化方法检测子宫内膜样腺癌组织中LRP16和E-cadherin表达部位及表达水平,分析两者之间的关系。结果子宫内膜样腺癌组织中LRP16mRNA的阳性表达率及表达水平均高于正常子宫内膜组织(P<0.05)。子宫内膜样腺癌组织中,LRP16核表达程度与病理学分级相关(P<0.05),而LRP16浆表达与病理学分级无相关性(P>0.05);LRP16浆表达与手术病理分期呈负相关(P<0.05),而LRP16核表达与手术病理分期无相关性(P>0.05);LRP16和E-cadherin的表达无相关性(P>0.05)。结论子宫内膜样腺癌组织中LRP16 mRNA的表达明显高于正常子宫内膜组织;且LRP16蛋白产物核表达水平与癌细胞的分化程度呈负相关;LRP16的过表达与子宫内膜样腺癌的发生和发展可能存在重要联系;而LRP16蛋白的表达水平与E-cadherin的表达可能无相关性。Objective Observe the expression of LRP16 in the tissues of endometrial endometrioid adenocarcinoma. In-vestgate the correlation between expression of LRP16 and E-cadherin in endometria cancer Methods RT-PCR was used to exam-ine the expression of LRP16 inendometrial endometrioid adenocarcinoma specimens. The immuno-histochemical method was used to examine the LRP16 protein in the tissue of endometrial endometrioid adenocarcinoma. Results The expression rate and level of LRP16 mRNA in endometrial cancer were higher than in normal tissue (P〈0. 05). LRP16 protein was expressed in the nucleus or cytoplasm of endometrial cancer specimens. The levels of LRP16 protein in the nucleus were related in the degrees of differentiation of the cancer cells (P〈0. 05). LRP16 expression in the cytoplasm was no related with grade (P〉0. 05). There was no relation-ship between LRP16 expression in the nucleue and staging(P〉0. 05). LRP16 expression in the cytoplasm was related with staging (P〈0. 05). No correlation was found between LRP16 and E-cadherin in cancer of endometrium. Conclusion The expression of LRP16 protein in the endometrial cancer is higher than that in the normal endometrial. The relationship between the degrees of differ-entiation of endometrial cancer cells and the expressions of endometrial protein is negative correlation. The over-expression of LRP16 should be related with the occurrence and development of endometrial cancer. But there was no significant difference between the ex-pression of LRP16 and E-cadherin in endometrial cancer.
