摘 要:目的探讨辛伐他汀对内毒素急性肺损伤大鼠肺组织中survivin表达水平的影响及其对减轻肺损伤的作用。方法注射内毒素制备SD大鼠急性肺损伤模型并随机分为LPS组、辛伐他汀组(Sta组)、辛伐他汀抑制组(Sta+Ly组)。分别于注射内毒素前(0 h)、注射后2、6、12及24 h时间点处死大鼠,应用RT-PCR、免疫组织化学等方法观察各组不同时间点肺内survivin表达水平的变化及肺组织损伤程度。同时设立注射生理盐水的对照组。结果与对照组相比,注射内毒素的大鼠肺组织损伤明显,且在各个时间点上肺内survivin表达水平均明显增加(P<0.05),且呈现注射内毒素后2 h出现表达,6 h升至顶峰,12 h逐渐下降的变化。与LPS组比较,Sta组survivin表达水平在各个时间点上均显著降低(P<0.05),而Sta+Ly组survivin表达水平虽较对应时间点的LPS组均有降低,但差异不具有显著性(P>0.05)。同时,与Sta组比较,Sta+Ly组survivin表达水平明显回升(P<0.05)。上述结果与组织切片观察的肺组织损伤程度的变化相一致。结论辛伐他汀可降低内毒素致急性肺损伤大鼠肺组织中survivin的表达水平,进而减轻内毒素致急性肺损伤程度。[Objective]To investigate the effect of Simvastatin on the expression of survivin of lung tissues in rats with acute lung injury(ALI) induced by endotoxin, and its role in alleviating the lung injury. [Methods]The SD rats were injected with endotoxin to induce ALI models, and then were randomly grouped into LPS group, Simvastatin group(Sta group) and Simvastatin inhibition group(Sta+Ly group). The expression of survivin in the lung tissues before injection(0 h) and 2, 6, 12 and 24 h after injection was analyzed by reverse transcription polymerase chain reaction(RT-PCR), immunohistochemistry assay, etc. Simultaneously, the rats were injected with normal saline in the control group. [Results]Compared to the control group, the lung injury of the rats injected with endotoxin was significant, the expression of survivin in the lung tissues of the rats with acute lung injure caused by endotoxin was significantly increased at each time point(P〈0.05). The level of survivin increased at 2 h, reached the peak at 6 h,and decreased at 12 h after ALI. Compared to those in the LPS group, the level of survivin in the Sta group was significantly decreased at each time point(P〈0.05); the level of survivin in the Sta+Ly group was also decreased at each time point, but without significant difference(P〉0.05). Compared to those in the Sta group, the level of survivin in the Sta+Ly group rebounded significantly(P〈0.05). The results were consistent with the results of the injury degree of the lung tissues by histological examination. [Conclusions]Simvastatin can decrease the expression of survivin of lung tissues in rats with acute lung injury caused by endotoxin and relieve the degree of injury.
