摘 要:目的检测便秘模型小鼠升、降结肠黏膜水通道蛋白4、9(AQP4、AQP9)mRNA表达情况,探讨AQP4、AQP9与便秘之间的关系。方法采用逆转录聚合酶链反应(RT-PCR)检测便秘组及对照组小鼠升、降结肠黏膜细胞AQP4 mRNA、AQP9 mRNA表达。结果对照组及便秘组小鼠升、降结肠均有AQP4 mRNA、AQP9 mRNA表达;便秘组升结肠中AQP4 mRNA表达量高于对照组,两组升结肠中AQP9 mRNA表达差异不显著;便秘组降结肠中AQP9 mRNA表达低于对照组,AQP4 mRNA表达高于对照组。结论便秘患者结肠AQP4 mRNA表达上调,AQP9 mRNA表达下调,AQP4、AQP9含量的增减会影响肠道水分的吸收与分泌,可能与便秘的发生、发展有一定的关系。Objective To investigate the expression of AQP 4 mRNA and AQP9 mRNA in colon mucosa of constipa-tion mice, and their roles in the constipation .Methods Semiquantitative reverse transcription-polymerase chain reac-tion ( RT-PCR) was used to detect the expression of AQP 4 mRNA and AQP9 mRNA in colonic mucosa .Results AQP4 mRNA and AQP9 mRNA expressed in ascending and decending colon of both normal and constipation groups; AQP4 mRNA expression was higher in ascending colon of constipation group than that of normal group , but AQP9 mRNA ex-pression in ascending colon was no significant difference between two groups;AQP9 mRNA expression was lower in de-scending colon of constipation group than that of normal group , but AQP4 mRNA was higher in descending colon of con-stipation group than that of normal group .Conclusion AQP4 mRNA increases in ascending and decending colon but AQP9 mRNA decreases in descending colon of constipation group .The increase or decrease of expression of AQP 4 mR-NA and AQP9 mRNA may be related with the occurrence and development of constipation .
