摘 要:目的:优化水提醇沉提取武当道茶茶多糖的方案并对茶多糖进行定量分析。方法:对多糖提取得率进行单因素试验,对提取次数、温度、料液比和时间进行L9(34)正交试验,采用蒽酮-硫酸法测定多糖含量。结果:以料液比1∶25在80℃下提取3次,每次提取4 h,茶多糖提取得率为7.49%,RSD=2.25%(n=3),多糖含量为4.28%,RSD=1.54%(n=5)。结论:上述方案多糖提取得率稳定,多糖含量高,可用于武当道茶茶多糖的开发和利用。Obieetive To optimize the method of water extraction and alcohol precipitation in extracting polysaccharide from Taoism Tea in Wudang mountains and quantitatively analyze the content of polysaccharide. Methods The extracting yield of polysaecharide was evaluated by a single factor experiment. Orthogonal tests were carried out on the four factors including the extracting times, extracting temperature, the ratio of material to water and extracting time according to L9 ( 34 ) scheme. The content of polysaceharide was determined by anthrone-sulphurie acid method. Results By using the method of extracting 3 times with 1 : 25 ratio of material to water 4 hours for each time at 80 ~C, the qualitative yield of polysaccharide extraction was 7.49%, RSD = 2.25% ( n = 3 ). The content of polysaeeharide in Taoism Tea in Wudang mountains was 4.28%, RSD = 1.54% ( n = 5). Conclusion The optimized method of water extraction and alcohol precipitation is suitable for polysac- charide extraction from Taoism Tea in Wudang mountains with the stable yield and high content, which would be used for its further development and utilization.
