摘 要:目的:西藏绿萝花乙醇提取物中,除乙酸乙酯和正丁醇萃取相外,石油醚萃取相也具有较强的α-葡萄糖苷酶抑制作用,该萃取相在降低餐后高血糖的同时是否影响糖尿病其他症状以及糖尿病肾病还值得进一步探讨。方法:本研究采用SD大鼠腹腔注射STZ并长期给予高蔗糖饲料,形成稳定的1型糖尿病模型,以50mg/kg、200mg/kg、500mg/kg剂量绿萝花提取物石油醚相高蔗糖饲料喂养,研究对糖尿病大鼠血糖、血脂、炎症因子、糖尿病肾病的影响。结果:研究发现,与模型组相比,50mg/kg绿萝花提取物石油醚相处理组即可使血糖、糖化血清蛋白、尿糖降低;甘油三酯及胆固醇水平也明显降低,且成剂量依赖性。血肌酐、血尿素氮、尿NAG酶显著下降,炎症介质IL-6的水平也明显降低。肾脏病理切片结果显示:与模型组相比,50mg/kg绿萝花提取物石油醚相处理组肾小球损伤减轻,肾小管肿胀好转,空泡减少。免疫组化结果显示:模型组大鼠肾脏组织染色阳性区域光密度值明显升高,TGF-β过度表达,50 mg/kg绿萝花提取物石油醚相处理组TGF-β表达降低,且成剂量依赖性。结论:绿萝花提取物石油醚相可通过抑制α-葡萄糖苷酶活性降低餐后血糖,且可以降低糖尿病大鼠的血脂、血肌酐、血尿素氮等,对肾脏起保护作用,其机制可能与抑制炎症因子IL-6以及TGF-β的表达有关。Objective: Except for ethyl acetate and N-butanol phase of Tibet Scindapsus aureus extracts,petroleum ether extract phase also have a strong inhibitory effect on α-glucosidase and decrease the postprandial hyperglycemia,Whether or not the phase have a beneficial effect on other symptoms of diabetes and diabetic nephropathy has not been reported. Methods: In this study,by intraperitoneal injection STZ and long-term high-sucrose diet for SD rats to formate a stable model of type 1 diabetes,taken 50 mg / kg,200 mg / kg,500 mg / kg different doses of Scindapsus aureus petroleum ether extract phase fed,to discover the drugs influence on blood glucose,blood lipids,inflammatory factors,diabetic nephropathy. Results: Compared with the model group,the 50 mg / kg Tibet Scindapsus aureus extracts petroleum ether phase treatment groups blood glucose,glucosylated serum protein,urine glucose decreased,as well as triglyceride and cholesterol. Moreover,serum creatinine,blood urea nitrogen,urinary NAG enzyme significantly decreased. The inflammatory mediators IL-6 is also decreaed obviously. Renal pathology results show that compared with the model group,the mice in the group treatment by 50 mg / kg Scindapsus aureus petroleum ether extract phase show symptoms of Glomerular injury mitigation,tubulus renalis swelling improvement and the number of cavitation reduce. The results of Immunohistochemistry show: In the model group of rats kidney tissue,optical density regions staining increased significantly,and TGF-β overexpressed. The dose of 50 mg / kg Tibet Scindapsus aureus extrac petroleum ether phase treatment decreased expression of TGF-β,and a dose-dependent manner. Conclusion: Tibet Scindapsus aureus extrac petroleum ether phase can reduce postprandial blood glucose by inhibiting α-glucosidase activity,and reduce blood lipids in diabetic rats,serum creatinine,blood urea nitrogen blood urea nitrogen to protect kidneys,the mechanism may be related to the inhibition of inflammatory cytokines expression of IL-6 and T
