摘 要:目的探讨抑制容量性钙内流(CCE)对胶质瘤U87细胞侵袭及迁移力的影响。方法采用免疫印迹(Western blot)检测基质相互作用分子1(STIM1)RNA干涉(RNAi)后U87细胞STIM1表达变化;活细胞钙成像检测CCE抑制剂(SKF-96365)处理及STIM1 RNAi后U87细胞CCE的改变;细胞划痕和Transwell侵袭实验检测CCE对U87细胞侵袭及迁移力的影响。结果 SKF-96365处理及STIM1RNAi均能明显抑制U87细胞的CCE(P<0.05);与对照组比较,抑制CCE后U87细胞迁移及侵袭能力明显降低(P<0.05)。结论抑制CCE可有效抑制U87细胞迁移及侵袭力,其机制可能与调控细胞内钙稳态相关。Objective The effects of capacitive calcium entry (CCE) inhibition on migration and invasion of glioma cell line U87 cells are discussed. Methods Stromal interaction molecule 1 (STIM1) expression of U87 cells treated with STIM1 RNA interfere (RNAi) were detected by Western blot. The cell cytosolic Ca^2+ concentration was measured by calcium imaging after SKF-96365 or STIM1 RNAi treatment. The migration and invasion ability were examined by in vitro wound healing assay and Transwell invasion assay, respectively. Results Calcium imaging showed SKF-96365 or STIM1 RNAi insults inhibited the CCE of U87 cells (P 〈0.05). Compared with control group, the migration and invasion of U87 cells treated with SKF-96365 or STIM1 RNAi were decreased obviously (P 〈0.05). Conclusion CCE inhibition suppressed the migration and invasion of U87 cells. The suppression might be associated with regulation of calcium homeostasis.
