摘  要:目的探讨骨髓间充质干细胞(BMSC)对血管性痴呆大鼠的保护作用。方法雄性SD大鼠随机分为正常组、模型组和移植组。采用双侧颈总动脉永久性结扎建立大鼠血管性痴呆(Va D)模型,模型建立24 h后,尾静脉移植第三代BMSC 3×106个。于移植结束后第14 d取海马组织Western blot分析缺氧诱导因子(HIF-1α),血管内皮生长因子(VEGF)和Caspase-3表达量的变化,免疫荧光分析血管密度的变化。结果移植组干预效果显著,VEGF表达量较模型组明显升高(P<0.05),Caspase-3表达量较模型组显著下降(P<0.05),且血管密度较模型组明显增高(P<0.05),而HIF-1α表达量则无明显差异(P>0.05)。结论骨髓间充质干细胞促进了血管生成,对血管性痴呆大鼠有一定的保护作用。Objective The neuroprotection of bone marrow mesenchymal stem cell (BMSC) on the bilateral common carotid artery occlusion model (BCCAO) is discussed.Methods Male Sprague-Dawley rats were randomly divided into three groups: normal group, vascular dementia (VaD) group and VaD-BMSC group. Rats in the VaDBMSC group were administered 3 × 10^6 BMSC via tail vein infusion at 24 h after BCCAO. The rats were sacrificed at 14 d after transplantation. Western blot analysis was used to analyze the expression of hypoxia-inducible factor-lot (HIF-1α), vascular endothelial growth factor (VEGF) and Caspase-3. Immunofluorescence analysis was used to analyze the bloed vessel density.Results Compared with the VaD group, the expression of VEGF in the VaD-BMSC group was significantly elevated (P 〈0.05), the expression of Caspase-3 in the VaD-BMSC group was significantly decreased (P 〈0. 05), the blood vessel density of VaD-BMSC group was significantly higher than that of VaD group (P 〈0. (15) and there was no significant difference of HIF-1α between the VaD group and VaD-BMSC group (P〉0.05).Conclusion BMSC could promote the angiogenesis and play a protective role on the rat model of vascular dementia.
