摘 要:目的:通过观察细胞周期蛋白激酶抑制因子p27在成年肌萎缩脊髓侧索硬化症(ALS)转基因鼠脊髓中的表达变化,探讨p27在ALS转基因鼠发病机制中的作用及其意义.方法:选取ALS转基因鼠和同窝野生型鼠,分别于95 d、108 d和122 d取材,分离脊髓,分别应用免疫荧光、免疫荧光双标记和免疫印迹,观察p27的分布和表达变化.结果:与同窝野生型鼠比较,p27蛋白在ALS转基因鼠发病早期(95 d)、中期(108 d)、晚期(122 d)均显著降低.免疫荧光实验结果显示,与同窝野生型鼠比较,ALS转基因鼠脊髓中p27阳性细胞数量减少,阳性细胞主要为p27/oligo-2双阳性细胞和p27/β-tubulinⅢ双阳性细胞,p27/GFAP双阳性细胞数量较少.结论:在ALS转基因鼠脊髓中p27阳性细胞数量减少,蛋白表达降低,表明p27与ALS的发病密切相关.Objective: To explore the expression change of cyclin-dependent kinase inhibitor p27 in the spinal cord of adult amyotrophic lateral sclerosis(ALS) transgenic mice, and to investigate the roles of p27 in the pathogenesis of ALS. Methods: Both the ALS transgenie mice and wild-type mice were selected at the age of 95 d, 108 d and 122 d. Their spinal cords were separated. Total proteins were obtained and observed by immunofluorescence technique and Western blotting. Results: Western blotting results showed that p27 expression was significantly decreased in the spinal cord at 95 d, 108 d, 122 d, compared with wild type mice. Immunofluorescence results showed that p27 positive cells were reduced in the spinal cord of ALS transgenic mice at different ages compared with litter born-wild type mice. Immunofluorescence staining showed that there were more p27/oligo-2 and p27/β-tuhulinⅢ double positive cells in the spinal cord of the ALS transgenic mice, but fewer p27/GFAP double positive cells. Conclusion= p27 positive cells and the expression of protein are both reduced in the spinal cord of ALS transgenic mice, suggesting that p27 is closely related to the pathogenesis of ALS.
