摘 要:目的探讨SDF-1/CXCR4生物轴对口腔鳞状细胞癌细胞生物学行为的影响。方法采用MTT法检测细胞的增殖能力。通过趋化实验观察细胞的侵袭迁移能力。结果口腔鳞状细胞癌细胞在SDF-1作用下增殖反应明显增强,CXCR4抗体可明显抑制肿瘤细胞的增殖(P<0.05)。趋化实验显示SDF-1可诱导口腔鳞状细胞癌细胞发生明显迁移,经50μg/m1CXCR4抗体孵育后,SDF-1介导Tca8113细胞的这种侵袭能力明显减弱(P<0.05)。结论 CXCR 4/SDF-1受体配体系统可介导口腔鳞状细胞癌细胞的增殖和迁移,在癌细胞向颈部淋巴结转移中发挥着重要作用。干扰SDF-1/CXCR4的相互作用有可能成为治疗口腔鳞状细胞癌的新方法。Objective To investigate the effect of chemokine SDF-1/CXCR4 biological axis on biological behavior of oral squamous cell carcinoma(OSCC). Methods Tea8113 ceils were cultured in RPMI-1640 medium containing 10% fetal bovine serum under 37℃ saturated humidity. MTT assay was used to evaluate the effect of SDF-1/CXCR4 on proliferation of the OSCC cells. The migration and cell invasion ability was detected by ehemotaxis assay. Results In MTT assay, recombinant SDF-1 stimulated proliferation of tumor cells and CXCR4 neutralization by monoclonal antibodies markedly inhibited cell proliferation ( P 〈 0.05 ). The ehemotactic migration of OSCC cells could be induced by SDF-1. This induction was significantly reduced by preincubating Tea8113 cells with 50μg /m1CXCR4mAb ( P 〈 0. 05 ). Conclusion CXCR 4/SDF-1 system may promote proliferation and migration of tumor ceils and may play an important role in lymph node metastasis of OSCC. Interfering in the interaction of SDF-1/CXCR4 can be a meaningful therapeutic target and strategy.
