摘 要:目的探讨C57BL/6.NOD-Aec1Aec2干燥综合征模型小鼠干眼体征严重程度及泪腺炎症反应与性别的关系。方法 C57BL/6.NOD-Aec1Aec2小鼠雌性和雄性组各45只,在生后第4、8、12、16、20周龄时进行泪液分泌实验(Schirmer’s testⅠ)、角膜荧光素染色及评分和泪腺组织HE染色、炎症细胞浸润计数评估及角膜上皮细胞表面微绒毛扫描电镜观察。结果第4周龄时,C57BL/6.NODAec1Aec2雌、雄性小鼠均未出现明显的干眼体征。从第8周龄开始,随着周龄增加,2组小鼠泪液分泌值均逐渐减少;角膜荧光素染色及评分值均明显加重;HE染色显示泪腺组织腺管、腺泡等结构破坏及炎症细胞浸润程度逐渐加重;扫描电镜观察显示角膜上皮细胞脱落增多,微绒毛减少,细胞紧密连接减少或中断。第16周龄时,雌性小鼠组较同龄雄性小鼠组的泪液分泌值显著减少(P=0.035),角膜荧光素染色评分高(P=0.028),雌性小鼠泪腺组织破坏及炎症细胞浸润程度更严重(P=0.017)。结论雌性C57BL/6.NOD-Aec1Aec2干燥综合征模型成年小鼠干眼体征较雄性更为严重,推测可能与雌激素水平变化有关。Objective To investigate the relationship of gender differences with severity of dry eye phenotype and histopathological changes of ocular surface in the C57BL/6. NOD-AeclAec2 mice (a mouse model of Sj6gren' s syndrome). Methods Totally 45 female and 45 male C57BL/6. NOD-AeclAec2mice were used in the study. Schirmer' s test I , corneal fluorescein staining and scoring, Hematoxylin-eosin (HE) staining, counting for inflammatory cells in the lacrimal gland, and scanning electron microscopy for corneal epithelial cells and microvilli were performed in postborn 4, 8, 12, 16 and 20 weeks. Results Both the female and male mice had no sign of dry eye at the age of 4 weeks. Since the 8th week, tear secretion became decreased which the stained grade of corneal fluorescein increased with the age growth in both gender groups. HE staining showed damaged structures of the ducts and acinar and severer infiltration of inflammatory cells in lacrimal glands. Scanning electron microscopy displayed dropped corneal epithelial cells, diminished microvilli and loosen or interrupted intercellular junctions. At the age of 16 weeks, the male mice had significantly decreased tear secretion ( P = 0. 035 ) . increased stained grade of corneal fluorescein (P =0. 028) and more inflammatory cells ( P = 0. 017 ) when Conclusion The signs of dry eye are more severe in the which may be related to the changes of estrogen levels. compared with the female mice at the same age. female C57BL/6.
