摘  要:目的检测血管内皮生长因子-C(VEGF-C)和环氧合酶-2(COX-2)在人宫颈癌细胞系中表达,探讨二者之间的关系及COX-2在VEGF-C介导的人宫颈癌细胞生物学行为中的作用。方法用Western blot方法检测VEGF-C和COX-2在HeLa、Siha、C33a和Caski等4株宫颈癌细胞中的表达;用特定浓度的COX-2特异性抑制剂NS398处理HeLa细胞,评估其对HeLa细胞的增殖、转移产生的影响。结果 VEGF-C和COX-2蛋白在4株宫颈癌细胞中均有表达,且在不同细胞株中二者表达强弱一致。用NS398处理HeLa细胞后,其VEGF-C蛋白表达明显降低,且细胞凋亡率增加。NS398可诱导HeLa细胞增殖活性降低(P<0.05),且呈剂量依赖模式。Transwell实验结果显示NS398处理组细胞侵袭力为(25.45±3.67)%,较对照组(40.38±0.72)%明显减弱(P<0.05)。结论 VEGF-C和COX-2均在人宫颈癌细胞中表达,且COX-2与VEGF-C表达之间有一定的相关性。COX-2可能是宫颈癌细胞中VEGF-C表达的一个调节者,其抑制剂NS398对于VEGF-C介导的肿瘤生长和转移有一定抑制作用。Objective To investigate the expressions of COX-2and VEGF-C,and their relationship in human cervical cancer,and to examine the role of COX-2in the VEGF-C mediated biological behaviors of human cervical cancer cells.Methods The expressions of COX-2and VEGF-C were detected in four human cervical cancer cell lines,HeLa,Siha,C33 aand Caski,by Western blot.The effects of COX-2specific inhibitor NS398 on the proliferation,apoptosis and migration of HeLa cells was evaluated after treatment with NS398.Results Both COX-2and VEGF-C were expressed in the four human cervical carcinoma cell lines and the expression levels of COX-2were coincident with those of VEGF-C in different cell lines.The expression of VEGF-C protein was significantly decreased and the apoptosis increased in HeLa cells treated with NS398.NS398 profoundly suppressed the proliferation of HeLa cells in a dose-dependent manner(P〈0.05).Transwell assay showed that the invasive ability was attenuated in NS398-treated cells as compared with control cells[(25.45±3.67)% vs.(40.38±0.72)%,P〈0.05].Conclusion Both VEGF-C and COX-2are expressed in human cervical cancer cells and there is an association between the expressions of COX-2and VEGF-C.COX-2 may be a regulator of VEGF-C and its specific inhibitor NS398 may inhibit VEGF-C mediated tumor growth and metastases in human cervical cancer.
