摘　　要：目的探讨转化生长因子β1(TGF-β1)对人宫颈癌He La细胞上皮-间质转化(EMT)发生的诱导作用。方法用10 ng/m L的TGF-β1处理He La细胞72 h,电镜下观察细胞形态,real-time PCR和Western blotting检测EMT相关上皮标记分子和间质标记分子的表达,划痕试验、Transwell实验检测细胞侵袭和转移能力。结果 TGF-β1可诱导He La细胞呈现EMT细胞形态,并且下调上皮相关标记蛋白E-cadherin和β-catenin的表达,上调间质相关标记蛋白Vimentin和N-cadherin的表达;细胞的侵袭和转移能力明显增强。结论 TGF-β1能够诱导He La细胞发生EMT,促进细胞侵袭和转移。Objective To investigate the effects of transforming growth factor β1（ TG F-β1） on inducing the epithelial-mesenchymal transition（ EM T） of H e La cells of human cervical cancer.Methods H e La cells were treated by TG F-β1of 10 ng/mL for 72 h and the morphological characteristics of H e La cell lines were observed under the electron microscope.The expressions of epithelial markers and mesenchymal markers relevant to EM Twere detected by the real-time PCR and W estern blotting.The invasion and migration of H e La cells were detected by the wound healing assay and Transwell chambers.Results TG F-β1could induce morphological alteration of H e La cells fromepithelial morphology to mesenchymal morphology, down-regulate the expressions of epithelial marker of E-cadherin and β-catenin, up-regulate the expressions of mesenchymal makers of V imentin and N-cadherin, and significantly increase the invasion and migration of H e La cells.Conclusion TG F-β1can induce the incidence of EM Tof H e La cells and enhance the invasion and migration of H e La cells.
