摘 要:目的固有免疫受体(Toll like receptors,TLRs)在鼻咽癌发病过程中起重要作用,该研究检测固有免疫受体9(Toll like receptor-9,TLR9)在鼻咽癌癌组织与癌旁组织中的表达,探讨信号因子TLR9表达与鼻咽癌临床病理之间的联系。方法收集近1年97例鼻咽癌患者临床标本,同时收集所有病例的一般情况和临床病理特征等资料。应用RT-PCR、Western blot方法检测TLR9在鼻咽癌及癌旁组织中的基因和蛋白表达水平。采用免疫组化、免疫荧光等方法检测TLR9在鼻咽癌及癌旁组织中的分布规律,并结合临床参数探讨TLR9与鼻咽癌临床病理特征的相关性。结果 RT-PCR结果显示,与癌旁组织相比,TLR9 m RNA在鼻咽癌组织中表达明显增高,具有显著统计学意义(P<0.01);Western blot结果显示,与癌旁组织相比,TLR9在鼻咽癌组织中表达明显增高,具有显著统计学意义(P<0.01)。TLR9蛋白表达在鼻咽癌组织中的增高与鼻咽癌的部分临床参数如肿瘤分化程度、临床分期等密切相关。免疫组化显示TLR9在癌旁组织中表达呈弱阳性,而在鼻咽癌组织中表达明显增强,其主要分布于肿瘤细胞以及炎性细胞等部位。免疫荧光进一步提示HLA-DR阳性的炎症细胞为TLR9因子分泌的重要来源。结论 TLR9在鼻咽癌组织中表达明显增高,且与临床参数密切相关,说明TLR9可能在鼻咽癌病理形成过程中发挥一定作用,可能作为判断临床预后的重要指标。To observe the distribution and expression of TLR9 (Toll like receptor 9) in nasopharyngeal carcinoma tissue and the adjacent normal tissue and explore the role of TLR9 in the nasopharyngeal carcinoma pathogenesis, clinical data of 97 cases of pathologically diagnosed nasopharyngeal carcinoma were collected and analyzed. RT-PCR and Western blot analysis were used to detect the expressions of TLR9 mRNA and protein levels in the nasopharyngeal carcinoma tissue and adjacent normal tissue. Immunohistoehemieal assay was employed to study the distribution characteristics of TLR9 in the nasopharyngeal carcinoma tissue and adjacent normal tissue. Furthermore, the relationship between TLR9 and nasopharyngeal carcinoma was evaluated. Data showed that the expression level of TLR9 mRNA was significantly increased in nasopharyngeal carcinoma as compared to adjacent normal tissues (P〈0.01). Western blot analysis further supported the similar results in protein expression of TLR9 in the nasopharyngeal carcinoma tissues (P 〈 0.01), further, immunohistochemistry demonstrated that TLR9 mainly distributed in tumor cells and inflammatory ceils. The expression levels of TLR9 closely related with the clinical parameters of tumor. TLR9 showed very weak expression in adjacent normal tissue. In conclusion, the gene and protein expression levels of TLR9 in nasopharyngeal carcinoma are significantly increased and closely associated with clinical progression, indicating that TLR9 plays a role in the development process of nasopharyngeal pathology, Thus TLR9 may be as an important prognostic indicator in clinical judgment.
