急性髓系白血病患者外周血细胞因子表达分析及临床意义

摘　　要：目的观察急性髓系白血病(AML)患者外周血白细胞介素-35(IL-35)、白细胞介素-10(IL-10)和转化生长因子-β(TGF-β)水平变化,并分析其临床意义。方法选取AML患者50例为研究组,选取同期50例健康体检的志愿者为对照组,清晨空腹采集所有AML患者及健康志愿者外周血,采用酶联免疫吸附测定法(ELISA法)检测2组样本血清IL-35、IL-10和TGF-β含量。结果与对照组比较,研究组患者血清IL-35和IL-10水平升高,TGF-β水平降低,差异具有统计学意义(P<0.05)。Pearson等级相关分析结果显示,研究组患者血清IL-10与TGF-β之间呈负相关,IL-35与IL-10之间呈正相关(P<0.05)。结论 AML患者血清IL-35和IL-10表达水平升高,TGF-β表达水平降低,且IL-35、IL-10和TGF-β之间密切相关,提示AML患者出现免疫状态异常,有助于AML的辅助诊断、疗效判断和发病机制研究。Objective To observe levels changes of serum cytokine interleukin -35（IL -35） , interleukin - 10（IL - 10） and transforming growth factor - β （ TGF - β ） in patients with acute myeloid leukemia （ AML）, and analyze the clinical value. Methods 50 patients with AML were selected as research group, and 50 healthy volunteers were studied as control group. Col- lecting the peripheral blood of all AML patients and healthy volunteers on empty stomach early in the morning. Enzyme - linked immunosorbent assay（ELISA）was used to detect the serum contents of IL -35, IL - 10 and TGF - β. Results Comparing with control group, the levels of serum IL - 35 and IL - 10 of research group increased, and that of TGF -β decreased, the difference was statistically significant（P 〈 0.05 ）. As a result of Spearman＇ test, the level of serum IL - 10 of research group was negatively correlated with TGF - β, and the level of IL - 35 was positively correlated with IL - 10 （P 〈 0.05 ）. Conclu- sion The levels of IL - 35 and IL - 10 of AML patients increased, and the TGF - β decreased. IL - 35, IL - 10 and TGF - β are closely related to each other, which indicate the abnormal immune status, it will contribute to the auxiliary diagnosis, effect of detection and pathogenesis in AML patients.
