摘  要:目的 观察纳米金(AuNPs)对荷肝癌小鼠电刀手术前后小鼠血清可溶性白细胞介素-2受体(sIL-2R)、转化生长因子-β(TGF-β)的影响.方法 实验分为不同浓度AuNPs处理组和生理盐水对照组,建立荷肝癌小鼠模型后,各组均经尾静脉注射0.2 ml相应的溶液,给药频率为2天1次,连续2周.然后经眶静脉取血,3d后电刀手术切取各组小鼠移植瘤,检测移植瘤重量和体积.手术后3周经眶静脉取血,采用酶联免疫吸附试验(ELISA)法检测电刀手术前后小鼠血清sIL-2R、TGF-β浓度变化.结果 手术前3d各处理组荷肝癌小鼠血清sIL-2R的浓度,空白对照组:(41.25 ±5.47) μg/L,低浓度AuNPs处理组:(25.53±2.73)μg/L,高浓度AuNPs处理组:(16.71±2.59)μg/L(P <0.01).手术后3周sIL-2R浓度,假手术组:(92.89±11.44) μg/L,单纯手术组:(19.97 ±4.27) μg/L,低浓度AuNPs+手术组:(8.59 ±2.83) μg/L,高浓度AuNPs+手术组:(8.38 ±5.79) μg/L(P <0.01).AuNPs对肝癌小鼠血清TGF-β的影响与上述结果类似.此外,不同浓度AuNPs处理组肝癌移植瘤重量及体积较空白对照组差异均有统计学意义(P<0.01),且呈剂量依赖关系.结论 电刀手术前后AuNPs处理均能够降低荷肝癌小鼠血清sIL-2R、TGF-β浓度,减小肝癌小鼠移植瘤重量和体积,发挥抗肿瘤作用.Objective To observe the influence of gold nanoparticles (AuNPs) on soluble interleukin-2 receptor (sIL-2R) and transforming growth factor-β (TGF-β) in hepatocellular carcinoma bearing mice before and after the electric knife treatment.Methods The experiment was divided into different concentrations of AuNPs-treated groups and the saline control group.After the establishment of hepatocellular carcinoma bearing mice model,0.2 ml solution in each group was injected via the tail vein every other day for 14 days.Then the transplanted tumors were taken out to measure the size and weight.The concentrations of serum sIL-2R and TGF-β in mice (3 days before and 3 weeks after the surgery) also were determined by enzyme linked immunosorbent assay (ELISA).Results (1) At 3rd day before surgery,the sIL-2R concentrations in control,2.5 and 5.0 mg/L AuNPs-treated groups were (41.25 ±5.47),(25.53 ± 2.73) and (16.71 ± 2.59) μg/L (P 〈 0.01).(2) At 3 rd week after the surgery,the sIL-2R concentrations in sham operation group,simple operation group,2.5 mg/L AuNPs + operation group and 5.0 mg/L AuNPs + operation group were (92.89 ± 11.44),(19.97 ± 4.27),(8.59 ± 2.83) and (8.38 ±5.79) μg/L respectively (P 〈 0.01).The effects of AuNPs on TGF-β concentration was similar to the above results.In addition,in AuNPs-treated groups,the xenograft weight and volume were significantly differenct from those in the control group in a dose-dependent manner (P 〈 0.01).Conclusion Gold nanoparticles can reduce serum sIL-2R and TGF-β concentrations before and after the electric knife treatment,also reduce hepatocellular carcinoma mouse xenograft weight and volume in vivo.
