摘 要:【目的】观察脐带间充质干细胞(umbilical cord mesenchymal stem cells,UCMSCs)的外分泌细胞因子联合亚低温疗法治疗脑损伤神经元的效果。【方法】经神经元裂解液诱导UCMSCs分泌的细胞因子,加入谷氨酸损伤的神经元培养基后进行常温或亚低温培养。神经元经谷氨酸损伤后,被随机分成4组:对照组、干细胞因子组、亚低温治疗组和联合治疗组。酶联免疫吸附法检测细胞因子浓度和乳酸脱氢酶(lactic acid dehydrogenase,LDH)含量,原位凋亡法观察细胞凋亡率。【结果】经神经元裂解液诱导后,UCMSCs条件培养液中神经生长因子(nerve growth factor,NGF)、脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)表达量明显增加,基质衍生因子-1α(stromal cell-derived factor-1α,SDF-1α)、转化生长因子-β-1(transforming growth factor-β-1,TGF-β-1)含量无明显变化(P>0.05)。条件培养液组、单纯低温治疗组和联合治疗组的LDH含量及细胞凋亡率均较对照组显著性降低(P<0.05);联合治疗组减轻神经元损失效果优于条件培养液组和单纯低温治疗组(P<0.05)。【结论】神经元裂解液诱导UCMSCs产生的外分泌细胞因子联合亚低温疗法对于减轻神经元损伤程度有明显作用。【Objective】To observe the therapeutic effects of exocrine cytokines of umbilical cord mesenchymal stem cells(UCMSCs)combined with mild hypothermia therapy on neuron injury.【Methods】Exocrine cytokines of UCMSCs induced by neuron lysate were added into the conditioned medium in which the neurons were damaged by glutamic acid, then cultured in normal temperature or mild hypothermia. Neurons damaged by glutamic acid were randomly divided into four groups: control group, conditioned medium group, mild hypothermia therapy group and combined therapy group. ELISA assay was used to determine the concentrations of exocrine cytokines and lactate dehydrogenase(LDH); apoptosis in situ assays were used to test the rate of cell apoptosis.【Results】The expression of nerve growth factor(NGF) and brain-derived neurotrophic factor(BDNF) in UCMSC conditioned medium increased obviously after the inducement of neuron lysate, but the expression of stromal cell-derived factor-1α(SDF-1α) and transforming growth factor-β-1(TGF-β-1) showed no significant change. The LDH level and cell apoptosis rate in conditioned medium group, mild hypothermia therapy group and combined therapy group were all significantly lower than those in control group. The effect reducing neuron injury in combined therapy group was better than that in conditioned medium group and mild hypothermia therapy group.【Conclusion】UCMSC exocrine cytokines induced by neuron lysate combined with mild hypothermia therapy have obvious effects on reducing neuron injury..
