摘 要:目的研究四妙勇安汤中绿原酸、甘草苷、阿魏酸的大鼠离体小肠吸收动力学。方法通过外翻肠囊法进行离体肠吸收实验,HPLC法测定不同取样时间肠内液中绿原酸、甘草苷、阿魏酸的含量并计算累计吸收量,以累计吸收量对时间作线性回归分析,计算吸收动力学参数。采用Kromasil 100-5 C18色谱柱(250mm×4.6 mm,4μm),流速:1.0 m L·min-1,柱温:25℃,其中绿原酸以乙腈-0.4%磷酸水为流动相按13:87比例洗脱,检测波长:327 nm;阿魏酸、甘草苷以乙腈(A)-1%醋酸(B)为流动相进行梯度洗脱(0-25min,80%-60%B),检测波长:245 nm。结果绿原酸吸收动力学方程为Y=0.937X-8.111,r=0.982;甘草苷吸收动力学方程为Y=0.150X-0.936,r=0.983;阿魏酸吸收动力学方程为Y=0.049X+1.448,r=0.995。结论四妙勇安汤中绿原酸、甘草苷和阿魏酸在大鼠离体小肠的吸收为一级动力学过程。Objective To determine the absorption kinetics of chlorogenic acid, licorice glycoside and ferulic acid in Simiao Yongan decoction in rat intestines. Methods In vitro intestinal absorption experiments were carried out by everting the gut sac. Chlorogenic acid, licorice glycoside and ferulic acid concentration were determined by HPLC. The concentration of cumulative absorption amount was calculated. The linear regression was figured through the concentration of cumulative absorption and time. Then the parameters of absorption kinetic were calculated. The column was Kromasil 100-5 C18(250 mm×4.6 mm, 4 μm). The flow rate was 1 m L·min^- 1. And the column temperature was 25 ℃. The mobile phase for chlorogenic acid consisted of acetonitrile-0.4% phosphoric acid(13 :87). The detection wavelength was 327 nm. The mobile phase for licorice glycoside and ferulic acid were acetonitrile and 0.1%phosphoric acid with gradient elution. The detection wavelength was 245 nm. The absorption site and mechanism of the 3 components in the rat small intestine were investigated.Results The regression equation of chlorogenic acid was Y = 0.937X- 8.111(r = 0.982), and the regression equation of licorice glycoside was Y = 0.150X- 0.936(r =0.983). The regression equation of ferulic acid was Y = 0.049 X + 1.448(r = 0.995). Conclusion The absorption of chlorogenic acid, licorice glycoside and ferulic acid in Simiao Yongan decoction conforms to the first-order kinetics in rats intestine.
