基因Ⅶ型新城疫病毒蛋鸡分离株的生物学特性及全基因组序列分析

摘  要:对广东某蛋鸡场新城疫病毒分离株(命名GD0526)进行生物学特性研究及全基因组序列分析,结果显示该分离株具血凝活性,能被特异性新城疫病毒标准阳性血清所抑制,动物回归试验出现新城疫典型症状。该毒株MDT为43.38h、ICPI为1.73、IVPI为2.37,F基因裂解位点为112R-R-Q-K-R-F117,符合强毒株特征。抗体增长规律和免疫交叉保护试验说明该毒株免疫原性较好,能产生较高水平的抗体,对试验鸡提供很好的保护。全基因组克隆和测序分析表明该毒株全基因组长15192bp,属基因Ⅶd型,与Shangdong-02-2010株核苷酸序列具有很高的同源性,而与LaSota株的相似性较低。The analysis of biological characteristics and the complete genome sequence of the Newcastle disease virus(NDV)isolated(named GD0526)from a layer farm in Guangdong,showed that GD0526 had HA activity and could be neutralized by the standard NDV positive serum. The animal regression experiment presented the typical symptoms of Newcastle disease. MDT,ICPI and IVPI of GD0526 were 43.38 h,1.73,2.37,respectively. GD0526 had the cleavage site(112R-R-Q-K-R-F117)in F gene,which was the symbol of a virulent strain. Antibody growth rule and cross-protection test showed that the isolate had a good immunogenicity which could stimulate the high level of antibodies to support immunity protection for chickens. The cloning and sequencing of the complete genome of GD0526 showed that the overall sequence was 15 192 bp and it belonged to genotype Ⅶ NDV which shared higher nucleotide sequence homology with Shangdong 02-2010 isolate,and less homology with the strain La Sota.
