摘  要:目的观察顺铂对人胃癌细胞系p53异构体Δ133p53、PTEN表达的影响。方法不同浓度顺铂作用于MKN45胃癌细胞系,采用MTT法检测细胞的抑制率;巢式逆转录多聚酶链反应(n RT-PCR)法检测p53异构体Δ133p53、PTEN m RNA水平的表达的变化。结果 MTT法检测结果示:MKN45胃癌细胞生长受到抑制,且抑制率随浓度、时间的增加而增加,与药物浓度为0μg/ml的对照组比较,顺铂能显著抑制MKN45胃癌细胞生长,且各组间差异均有统计学意义(P<0.01)。n RTPCR法检测结果显示:随着药物浓度增加,Δ133p53 m RNA表达呈减少趋势,PTEN m RNA表达显著增加。结论在顺铂诱导的人胃癌细胞系MKN45生长抑制实验中,PTEN表达增强可能是细胞生长抑制的重要机制之一,Δ133p53异构体表达减弱可能是PTEN表达增强的重要因素。Objective To observe the effect of cisplatin on the expression of PTEN andΔ133p53 isoform in MKN45 gastric cancer cell line.Methods Different concentrations of cisplatin were added to MKN45 gastric cancer cells.The inhibition rate was detected by MTT.The expression changes of PTEN andΔ133p53 mRNA were measured by nested reverse transcriptase polymerase chain reaction(nRT-PCR). Results MTT test results showed that the growth of MKN45 gastric cancer cells was inhibited by cisplatin, and the inhibition rate increased correspondingly with the concentration of cisplatin and acting time. Compared with the control group,different concentrations of cisplatin could significantly inhibit the growth of MKN45 cancer cells,and the differences were statistically significant(P〈0.01).In nRT-PCR analysis,the result showed thatΔ133p53 mRNA decreased,while PTEN mRNA increased significantly with the increase of drug concentration.Conclusions In this cisplatin-induced MKN45 inhibition experiment, PTEN plays a pivotal role,which may be regulated by a special p53 isoform-Δ133p53.
