微小染色体维持蛋白5基因在痰液脱落细胞表达对肺癌的诊断价值

摘　　要：目的:探讨微小染色体维持蛋白5(MCM 5)在痰脱落细胞中的表达,用以检测肺癌细胞,为临床早期发现、探讨方便、快捷的方法。方法:收集肺癌病人和健康志愿者痰脱落细胞,提取总RNA,逆转录-巢式聚合酶链反应,免疫组化检测微小染色体维持蛋白5 mRNA表达水平。结果:在肺癌36例痰脱落细胞中MCM 5阳性率58.3%,肺部良性疾病26例MCM 5阳性率15%,健康志愿者20名MCM 5阳性率0%。肺癌组织中MCM 5基因表达量在1、2、3级间差异均有非常显著意义(P<0.001)。结论:痰液脱落细胞中MCM 5基因检测有助于肺癌的临床诊断,对早期诊断亦有一定的意义。可以设想作为早期诊断肺癌的标志基因。Objective：To study the expression of MCM 5 gene in sputum cells of patients with lung cancer in order to research the diagnosis. Methods：We collected the sputum samples from 36 cases of lung cancer and 26 cases of benign pulmonary disease,extracted the total RNA,and then did RT-nest PCR,immunohistology to check the expressive level of mcm 5 gene. Results：Sensitivities of that are 58. 3%in 36 lung cancer,15% in other patients and 0%in normal person. The expression of MCM 5 between G1、G2 and G3 lung cancer tissue have very marked statistic significance （ P〈0 . 01 ） . Conclusion：It is usable to test the ex pressive level of MCM 5 gene in lung cancer to diagnosis and prediction.
