摘  要:目的探讨大鼠创伤后应激障碍(PTSD)易感性差异的腹侧被盖区(VTA)多巴胺D2受体(D2R)和多巴胺转运体(DAT)机制。方法 150只雄性Sprague-Dawley大鼠随机分为试验组120只和对照组30只。实验组建立PTSD模型,根据旷场试验(OFT)和强迫游泳试验(FST)测评指标将大鼠分为高、中、低易感组。在应激模型建立后3 h、4周和6周分别处死高、低易感组和对照组大鼠各8只,原位杂交检测大鼠VTA区D2R和DAT的表达。结果高易感组VTA区DAT的灰度值在三个时点高于低易感组,且差异均有统计学意义(P<0.01);高易感组D2R的灰度值在三个时点均低于低易感组,且差异有统计学意义(P<0.01)。结论 VTA区DAT的低表达和D2R高表达状态与大鼠PTSD高易感性有关。Objective To explore the expression of dopamine transporter(DAT)and dopamine D2 receptor(D2R)located in ventral tegmental area(VTA)in high and low PTSD susceptibility rats,and to unveil the possible mechanism lead to different susceptibility to post trau-matic stress disorder(PTSD). Methods One hundred and fifty male Sprague - Dawley rats were randomly assigned to experimental group(n =120)and control group(n = 30). Rats in experimental group were trained according to establish the procedure of PTSD. Open Field Test(OFT) and Forced Swimming Test(FST)were used to assess different susceptibility to PTSD:high susceptibility group(H group)and low susceptibility group(L group),each group accounted for 20% of total rats. After these models had been established,rats were scarified and cardio - perfused, their brains were removed and coronarily sliced up and VTA was selected. Levels of mRNA of DAT and D2R were estimated with in situ hybridiza-tion. Results The expression of DAT in H group was significantly lower than that of control and L groups in three time points( P 〈 0. 01),and the difference between H and L groups was also significant( P 〈 0. 01). However,the expression of D2R in H group was significantly higher than that of control and L groups in three time points,and the difference between H and L groups was significant( P 〈 0. 01). Conclusion The lower expression of DAT and high regulation of D2R in VTA may play an important role in rats with high susceptibility to PTSD.
