摘 要:目的:探讨乳腺癌新辅助化疗(NAC)前后雌激素受体(ER)、孕激素受体(PR)、人表皮生长因子受体(HER2)和拓扑异构酶Ⅱα(TOPOⅡα)蛋白表达的变化及其与疗效病理评价的关系。方法:收集80例经NAC的乳腺癌化疗前、后病理标本,采用免疫组化方法检测乳腺癌组织中ER、PR、HER2和TOPOⅡα蛋白表达的变化,并观察疗效病理评价。结果:NAC前、后HER2和TOPOⅡα蛋白的表达改变差异有统计学意义(P=0.008,0.006),ER和PR蛋白的表达改变差异无统计学意义(P=0.853);化疗疗效病理评价与NAC前后ER、HER2和TOPOⅡα蛋白的变化呈正相关关系(r=0.430,0.415,0.507,P=0.033,0.02,>0.001),而与PR蛋白的变化无相关关系(r=-0.122,P=0.308)。结论:NAC使乳腺癌组织的分子标记物ER、PR、HER2和TOPOⅡα蛋白发生了变化,并且与乳腺癌的治疗疗效有一定的关系,尤其是ER、HER2和TOPOⅡα蛋白表达的变化有重要意义。Objective:To investigate the changes of estrogen receptor (ER) ,progesterone receptor (PR) ,hu- man e ers of b dermal growth factor receptor (HER2) and topoisomerase Ⅱα (TOPO Ⅱα) proteins these biomark- reast cancer patients after treated with neoadjuvant chemotherapy(NAC). Methods:80 cases breast cancer samples treated with NAC and their match control samples were selected and used to detect the expression of ER, PR, HER2 and TOPO Ⅱα protein by immunohistochemistry,so that investigating the correlation with the NAC therapeutic effect. Results: There were significant changes in the HER2, TOPO IIaexpression after treated with NAC. ( P =0. 008 and 0. 006). However, there was no significant changes in the ER expression and PR expression after NAC. ( P =0. 853). Furthermore, there was a positive corre- lation between NAC chemotherapy efficacy and the changes of ER, HER2 and TOPO Ⅱα proteins. Conclu- sion:The changes of biological markers ER, HER2 and TOPO Ⅱα proteins were positive correlated with the NAC efficacy, these biomarker can be used as a potential prognostic indicators.
