ERCC1和TUBB3在晚期胃癌组织中的表达及其临床意义

摘　　要：目的通过检测核苷酸切除修复交叉互补组1(excision repair cross complementation group 1,ERCC1)和3型β微管蛋白(β-tubulin-3,TUBB3)在晚期胃癌患者肿瘤组织中的表达,并探讨两者相关性及其临床意义。方法采用免疫组化法检测手术切除的66例晚期胃癌组织及40例癌旁组织中的ERCC1及TUBB3蛋白的表达情况,并结合临床病理资料综合分析。结果ERCC1和TUBB3在晚期胃癌组织中的高表达率分别为59.1%和13.6%,在癌旁正常组织中的高表达率分别为30%和72.5%,两种蛋白高表达率在晚期胃癌组织和正常胃组织间差异均有统计学意义(P<0.05)。ERCC1、TUBB3蛋白表达与性别、年龄、肿瘤大小、浸润深度、淋巴结转移、肿瘤原发部位均无关(P>0.05),但与肿瘤的分化程度相关(P<0.05),ERCC1与TUBB3的表达成负相关。结论在晚期胃癌患者肿瘤组织中ERCC1成高表达、TUBB3呈低表达,两者在癌组织中表达具有密切关系。Objectve Through investigation on the excision repair cross complementation group-1（ERCC1）and β-tubulin-3（TUBB3） in advanced gastric carcinoma to explore their roles and clinical significance in the development of advanced gastric carcinoma.Meth-ods The expression of ERCC1 and TUBB3 in 66 resected patients with advanced gastric carcinoma and corresponding 40 normal tis-sues were detected by the method of immunohistochemistry.Results The high expression rates of ERCC1 and TUBB3 were 59.1%and 13.6% in advanced gastric carcinoma,respectively,and both were higher than those in normal tissues in which the high expression rates were 30%and 72.5%,respectively.The expression of both ERCC1 and TUBB3 showed a significant difference from those in nor-mal gastric tissues（P〈0.05）.No difference of ERCC1 or TUBB3 protein was found among patients with different genders,ages,tumor size,invasion depth,lymph node metastasis and tumor site （P〉0.05）,but all related to degrees of differentiation,and there is a nega-tive correlation between ERCC1 and TUBB3.Conclusions In advanced gastric carcinoma,ERCC1 shows high expression rates and TUBB3 shows low expression rates.There is also a negative correlation between ERCC1 and TUBB3 protein expression.
