宫颈癌乏氧诱导因子-1α和谷胱甘肽-S-转酶-π的表达及与化疗药物耐药性关系的研究  被引量:5

摘  要:目的观察乏氧诱导因子-1α(HIF-1α)和谷胱甘肽-S-转酶-π(GST-π)在宫颈癌中的表达,探讨其与化疗药物敏感性的关系。方法应用MTT法对71例未经化疗的宫颈癌患者行体外化疗药物阿霉素、卡铂、紫杉醇5-氟尿嘧啶和顺铂的敏感性检测,并用免疫组织化学方法对HIF-1α和GST-π在宫颈癌中的表达进行检测,将其表达情况与对应的化疗耐药性进行相关分析。结果 71例宫颈癌患者对紫杉醇(TAXOL)、顺铂(DDP)、阿霉素(ADM)、5-氟尿嘧啶(5-Fu)和卡铂(CBP)的耐药率分别为38.03%(27例)、53.50%(38例)、54.93%(39例)、26.80%(19例)和47.90%(34例)。腺癌耐药例数多于鳞癌,但两者差异无统计学意义(P>0.05)。HIF-1α和GST-π在71例宫颈癌患者中阳性率分别为56.34%(40/71)和57.75%(41/71),HIF-1α和GST-π在腺癌中的阳性率高于鳞癌,但两者差异无统计学意义(P>0.05)。HIF-1α和GST-π在宫颈癌中的表达呈明显相关,P<0.05。HIF-1α和GST-π的表达与化疗药物敏感性之间均存在明显相关(P<0.05)。结论 HIF-1α和GST-π可能共同参与介导宫颈癌的多药耐药,两者在宫颈癌中的阳性表达及共表达可作为预测宫颈癌患者多药耐药的指标之一。【Objective】To observe the expressions of hypoxia inducible factor 1α(HIF-1α) and glutathione S transferase π(GST-π) in cervical carcinoma and investigate the relationship between the expressions and the drug sensitivity. 【Methods】 Seventy-one cases of cervical carcinoma were tested with MTT assay to evaluate their chemoresistance to taxinol, cisplatin, carboplatin, adriacin and 5-fluorouracil. The expressions of HIF-1α and GST-π were examined with the immunohistochemical assay, and then the relationship between the expressions of the two proteins mentioned above and the chemoresistance was analysed. 【Results】The drug resistance rate of taxinol, cisplatin, adriacin, 5-fluorouracil and carboplatin in cervical carcinoma were 38.03%(n = 27), 53.50%(n = 38),54.93%(n = 39), 26.80%(n = 19) and 47.90%(n = 34), respectively. The cases of drug resistance of adenocarcinoma were higher than those of squamous carcinoma, but there was no significant difference between them(P 〉0.05).The positive rates of HIF-1αand GST-π were 56.34%(40/71) and 57.75%(41/71), respectively. The positive rate of HIF-1α and GST-π in adenocarcinoma was higher than that in squamous carcinoma, but there was no significant difference between them(P〉 0.05). The expression of HIF-1α was significantly correlated with the expression of GST-π in cervical carcinoma(P〈0.05). The positive expressions of HIF-1α and GST-π were significantly correlated with chemoresistance(P〈0.05). 【Conclusions】HIF-1α and GST-π may co-work to intervene the multidrug resistance(MDR) of cervical carcinoma, and the positive expression of them may be one of the index of forecasting MDR of cervical carcinoma patients.
