CIP2A在卵巢癌组织中的表达及其临床意义研究

摘　　要：目的探讨CIP2A在卵巢癌组织中的表达情况和临床意义。方法选取2008年1月至2010年7月在中国医科大学附属盛京医院手术切除的卵巢癌组织77例及正常卵巢组织6例,并收集其临床病理特征。采用兔抗人CIP2A多克隆抗体以SP免疫组化法、实时定量PCR进行研究,并分析CIP2A表达与临床病理因素的关系。结果在6例正常卵巢组织中,CIP2A的表达为阴性或低表达;而在77例卵巢癌组织中,33.77%(26/77)具有CIP2A阳性表达。CIP2A在卵巢浆液性癌、子宫内膜样癌、黏液性癌和透明细胞癌中的阳性表达率分别为39.13%(18/46)、23.53%(4/17)、16.67%(2/12)和100%(2/2)。CIP2A的表达与卵巢癌病理分期和肿瘤病理分级有关。实时定量PCR结果证实,相比正常卵巢组织,卵巢癌组织CIP2A转录水平显著提高(P<0.05)。结论 CIP2A在卵巢癌中高表达并且与FIGO分期和肿瘤病理分级有关。CIP2A可能成为卵巢癌诊断的新指标和治疗中的新靶点。Objective To investigate expression and clinical significance of C1P2A in ovarian cancer tissues. Methods CIP2A expression was analyzed in 77 ovarian cancer specimens in Shengjing Hospital of China Medical Uni- versity between January 2008 to July 2010 and their clinicopathologic factors were achieved. The expression of CIP2A and its correlation with elinicopathologic factors were analyzed by using rabbit anti-human polyclonal antibody with SP immunohistochemistry and RT-PCR.Results The expression of CIP2A in 6 normal ovarian tissues was negative or with low expression. CIP2A was overexpressed in 33.77% （26/77） cases of ovarian cancer. The positive overexpression rates of CIP2A in serous carcinoma, endometrioid carcinoma, mucinous carcinoma and clear cell carcinoma were 39.13% （18146）, 23.53% （4/17）, 16.67% （2/12） and 100% （2/2） respectively. Further studies showed that CIP2A overexpression positively correlated with advanced FIGO stage and tumor grade）. RT-PCR results showed that the tran- scriptional level of CIP2A increased remarkably in ovarian caner tissues compared with normal ovarian tissues （P〈0.05）. Conclusions These results validate that CIP2A is highly expressed in ovarian cancers and positively corre- lated with advanced FIGO stage and tumor grade.CIP2A is established as a promising oncogene of diagnosis and thera- peutic target of ovarian cancer.
