摘 要:目的探讨一种快速、有效提取黄芩中四种主要黄酮类物质的离子液体微波辅助法。方法在确立了黄芩中四种主要黄酮类物质的高效液相(HPLC)检测方法的基础上,通过单因素实验对提取时间、微波功率、离子液体浓度、固液比、提取次数五个因素进行了优化,确立了黄芩中四种主要黄酮类物质的提取工艺,并与药典法进行了比较。结果最优条件为,以1 mol/L的[C8mim]Br水溶液为提取溶剂,固液比1∶6(g/ml),在400 W、80℃的微波条件下提取90 s,提取1次。将优化后的提取方法与药典提取法进行比较,发现前者提取时间由3 h缩短到90 s,且对四种主要黄酮类物质提取率均略有提高。结论通过对提取时间、微波功率、[C8mim]Br浓度、固液比、提取次数五个主要影响因素的优化,微波辅助[C8mim]Br水溶液法可快速、有效地提取黄芩中黄酮类物质。Objective To establish a fast and efficient method for the simultaneous extraction of baicalin,wogonoside,baicalein and wogonin from Scutellaria baicalensis Georgi by optimizing the parameters of [C8mim]Br based microwave-assisted extraction([C8mim]Br MAE). Methods On the basis of the HPLC method for the four flavonoids,the extraction time,the microwave power,the concentration of [C8mim]Br,the solid-liquid ratio and the number of extraction cycles were optimized. Then the results by the proposed[C8mim]Br MAE approach was compared with that by pharmacopoeia method. Results The optimum [C8mim]Br MAE conditions were found to be solid- liquid ratio of 1∶ 6( g / ml),irradiation power of 400 W,irradiation time of 90 s,[C8min]Br concentration of1. 0 mol / L and one extraction cycle. The proposed approach provided higher extraction yield and less time( from 3 h to 90 s) than that of traditional pharmacopoeia extraction method. Conclusion The proposed [C8mim]Br MAE approach is proved to be a fast and efficient method for the extraction of baicalin,wogonoside,baicalein and wogonin from S. baicalensis.
