摘 要:以金鲳鱼内脏为超氧化物歧化酶(SOD)提取原料,优化提取因素,主要考察了提取物料比、温度、时间、p H、缓冲液浓度,在单因素实验基础上,采用正交实验,优化SOD提取工艺。通过正交实验获得SOD最佳提取工艺,物料比1:4,提取温度40℃,提取液p H6.4,Tris-HCl缓冲液浓度0.1 mol/L,提取时间4 h,在此条件下每克物料提取SOD活力944 U。实验采用鱼内脏提取海洋生物源SOD,获得更为安全高效的海洋生物源自由基清除剂产品,同时减少因加工鱼产品产生大量下脚料所带来的环境污染及资源浪费,实现废物利用,变废为宝。The object of this research was to optimize the superoxide dismutase(SOD) extraction technology from fish viscera by orthogonal test and single factor experiment, mostly investigated the influence of material ratio, extraction temperature, time, p H and buffer concentration. According to orthogonal experiments, the optimum process condition were as follows: material ratio was 1:4, extract for 4 hours at 40 ℃ with 0.1 mol/L Tris-HCl buffer solution, and p H 6.4, the SOD activity was 944 U/g fi sh viscera under optimum process conditions. The fi sh viscera was used as the raw material of SOD to obtain safe and effi cient free radical scavengers from marine sources, in addition, the technology can reduce the environment pollution and the waste of resources due to the leftover material in the aquatic production processing, which change the waste into valuables.
