摘 要:目的对牛奶中硫氰酸盐离子色谱测定法的样品预处理方法和色谱条件进行改进,提高方法的适用性。方法牛奶样品用氢氧化钠溶液加热沉淀蛋白,以AS14阴离子交换柱分离,3.5 mmol/LNa2CO3~1.0 mmol/L Na HCO3溶液洗脱,电导检测器检测。结果硫氰酸盐浓度在0.1mg/L^2.00 mg/L范围内线性良好,回归方程y=1.314×10-6x+0.024,r=0.9997,最低检出浓度0.8 mg/kg,精密度为1.5%~3.6%、加标回收率为93.5%~101.6%。结论本方法简便,快速,灵敏,适用于牛奶中的硫氰酸盐的检测。Objective To enhance the sample pretreament method and chromatographic condition for improvement of the thiocyanate in milk by ion chromatography. Methods The proteins in milk were precipitated by heating sodium hydroxide solution. Thiocyanate was separated with AS14 anion exchange column, eluted with 3.5 mmol/L Na2CO3-1.0 mmol/L NaHCO3, and quantified with conductivity detector. Results The linearity range of this method was from 0.1 mg/L-2.00 mg/L. The regression equation was y=1.314 × 10^-6x+0.024, r=0.9997. The detection limit was 0.8 mg/kg, with the precision of 1.5%-3.6%. The extraction recoveries with the method were 93.5%-101.6%. Conclusion This method is simple, rapid and sensitive. Besides, it is suitable to determine the thiocyanate in milk.
