摘　　要：目的研究抗坏血酸对成骨细胞膜片BMP-2mRNA表达的影响.方法：原代培养并鉴定大鼠成骨细胞,采用物理刮取法,体外构建成骨细胞膜片;成骨细胞接种后,分别加入15、50、85mg/L抗坏血酸,连续培养1～2周,RT-qPCR检测成骨细胞膜片BMP-2mRNA的表达.结果：获得的细胞符合成骨细胞形态学特征特性.可构建管状形态的成骨细胞膜片.组间比较,第1周和第2周成骨细胞膜片中BMP-2mRNA表达量实验组均高于对照组,以50mg/L组的表达量最高（第1周P〈0.05;第2周P〉0.05）;组内比较,各组表达量第2周均高于第1周（P〈0.05）.结论：抗坏血酸可促进成骨细胞膜片BMP-2mRNA表达.Objective： To study the effects of ascorbic acid on BMP-2 mRNA expression of osteoblast cell sheets. Methods： Rat os- teoblasts were primarily cultured and identified; osteoblast cell sheets were built by physical scraping method in vitro; the osteoblast cell sheets were cultured with 15, 50 and 85 mg/L ascorbic acid for 1 and 2 weeks respectively, and the expression of BMP-2 mRNA of the cell sheets was detected by RT-qPCR. Results： The obtained cells were conformed to be osteoblasts. The osteoblast cell sheets could be rolled into tube i1~ vitro. The expression of BMP-2 mRNA of osteoblast cell sheets in experiment group, whether in week one or week two was higher than that in control group, 50 mg/L group showed the highest expression （ first week P 〈 0.05 ; second week P 〉 0.05 ） ; the expression of any group in week two was higher than that in week one （P 〈 0.05 ）. Conclusion： Ascorbic acid may pro- mote the expression of BMP-2 mRNA in osteoblast cell sheets.
