摘 要:目的:近年来的研究表明,高迁移率族蛋白(1HMGB1)在肿瘤的发生及恶性演变过程中发挥重要作用,本研究旨在探讨HMGB1在胃癌组织、正常组织、胃癌细胞系SGC-7901、BGC-823、HGC-27、AGS及正常胃黏膜细胞系GES中表达情况。方法:免疫组织化学法检测HMGB1在32例可手术切除的胃癌患者组织标本(包括癌组织和正常组织)的表达情况;RT-PCR及Westren Blot检测HMGB1在胃癌细胞系SGC-7901、BGC-823、HGC-27、AGS及正常胃粘膜细胞系GES的m RNA及蛋白质表达。结果:胃癌组织HMGB1免疫组织化学染色评分高于正常组织(P<0.05);RT-PCR结果显示SGC-7901、BGC-823、HGC-27、AGS、GES细胞系HMGB1 m RNA表达丰度均较高;Westren Blot检测发现胃癌细胞系SGC-7901、BGC-823、HGC-27中HMGB1蛋白水平显著高于胃癌细胞系AGS及正常胃粘膜细胞系GES。结论:HMGB1在胃癌组织及正常组织中的表达具有显著性差异。胃癌细胞系SGC-7901、BGC-823、HGC-27相对于其它胃细胞系存在HMGB1高表达,适合后续基因敲除分析工作。Objective: Recent studies suggest that HMGB1 plays an important role in tumorigenesis and carcinoma malignant transformation. This study aims to examine the expression of HMGB1 in gastric cancer tissues and normal tissues, gastric cancer cell lines(SGC-7901, BGC-823, HGC-27, AGS) and human gastric mucosa epithelium cell line(GES). Methods: Immunohistochemistry staining was used to assess HMGB1 expression in 32 cancerous tissues and normal tissues from the resected gastric samples. RT-PCR and Western-blot were performed to determine HMGB1 m RNA level and protein level in gastric cancer cells, including SGC-7901,BGC-823, HGC-27, AGS and GES. Results: The overexpression of HMGB1 was found in cancerous tissues compared with normal tissues(P〈0.05). Furthermore, we found that the m RNA level of HMGB1 was all high in SGC-7901, BGC-823, HGC-27, GES and AGS measured by RT-PCR. In addition, we also found that the protein level of HMGB1 was higher in SGC-7901, BGC-823, HGC-27 than GES and AGS measured by Western blot. Conclusions: The expression of HMGB1 is significantly different between gastric cancerous tissues and normal tissues. The expression of HMGB1 are higher in SGC-7901, BGC-823 and HGC-27 than the other gastric cell lines,thus these cell lines are suitable for HMGB1 gene knock down analysis in the subsequent work.
