摘 要:目的探讨氢饱和生理盐水(hydrogen rich saline,HRS)对内毒素诱导的大鼠葡萄膜炎(endotoxin-induced uveitis,EIU)是否有减轻作用。方法健康成年雄性SD大鼠按随机数字表法分为正常组、模型组、氢饱和生理盐水(氢水)组和激素组,于后3组大鼠足垫部皮下注射0.125 mg/kg内毒素诱导葡萄膜炎。氢饱和生理盐水组大鼠于造模后0、0.5、1、2、6、8、12、24 h和3周内每天1次,腹腔注射氢饱和生理盐水20 m L/kg;激素组大鼠于造模后立即腹腔注射地塞米松磷酸钠注射液1 mg/kg。4组于造模后12、24、48、72、96 h在裂隙灯下观察葡萄膜炎表现,并进行量化评分;并于造模后1、4、7、10、14 d和21 d行视网膜电图检查、房水蛋白定量及病理组织学检查。结果氢饱和生理盐水组葡萄膜炎程度与模型组相似,裂隙灯临床表现量化评分和炎症细胞计数差异均无统计学意义(P>0.05);内毒素诱导的大鼠葡萄膜炎模型的视网膜功能有改变,暗适应3.0反应b潜伏期延长;与模型组相比,氢饱和生理盐水组b波潜伏期有缩短趋势,但差异无统计学意义(P>0.05);氢饱和生理盐水组房水蛋白总量较模型组略低,差异有统计学意义(P<0.05)。结论氢饱和生理盐水对内毒素诱导的大鼠葡萄膜炎无明显减轻作用。Objective To investigate whether hydrogen rich saline (HRS) can ameliorate endotoxin- induced uveitis (EIU) in rats. Methods Healthy adult male SD rats were randomly divided into a normal group, a model group, an HRS group, and a dexamethasone the later 3 groups by subcutaneously injecting 0. 125 mg/kg (DE) group, and EIU was induced in the rats of of hpopolysaccharide (LPS) into the footpads. The rats of the HRS group were injected with 20 mL/kg of HRS intraperitoneally at time points of 0, 0. 5, 1, 2, 6, 8, 12, and 24 h and once a day, for 3 weeks after injection of LPS. The rats of the DE group were injected with 1 mg/kg of DE sodium phosphate injection intraperitoneally and immediately after injection of LPS. All the rats were examined under a slit-slam microscope and graded according to the clinical signs of uveitis at time points of 12, 24, 48, 72, and 96 h after injection of LPS. Electroretinogram (ERG), aqueous humor protein quantification and histological examination were carried out on 1, 4, 7, 10, 14 and 21 d after injection of LPS. Results The rats in the HRS group and the model group showed similar uveitis symptoms, and there were no significant differences in the slit-lamp microscope quantitative scores and inflammation cell counts (P 〉 0. 05 ). Retinal function of the EIU rats was changed, with an elongated b wave potential in dark adaption 3.0 reaction. Compared with the model group, the b wave potential of the HRS group was shortened to some extent, but with no significant difference (P 〉 0. 05 ). Aqueous humor protein level of the HRS group was slightly lower than that of the model group, with statistically significant difference (P 〈 0. 05 ). Conclusion HRS cannot significantly ameliorate EIU in the rats.
