摘  要:目的研究槲皮素对肝癌高转移细胞基质粘附能力的影响及其机制.方法研究对象为人高转移肝细胞癌细胞HCCLM6,用细胞基质粘附实验检测槲皮素对细胞外基质粘附能力的影响,免疫荧光和Western blot检测细胞内E-Cadherin蛋白表达.结果槲皮素能抑制肝癌细胞对Marigel胶的粘附,免疫荧光和Western blot检测均发现槲皮素处理组肝癌细胞E-Cadherin蛋白表达较对照组增加(P<0.05).结论槲皮素能抑制肝癌高转移细胞的基质粘附能力,其机制之一可能在于槲皮素能上调肝癌细胞E-Cadherin蛋白表达.Objective To investigate the influence of quercetin on the adhesive ability of high-metastatic hepatoma cells to extraeeflular matrix and its mechanism. Mehtods Human high -metabasis HCC cell line (HCCLM6) was taken as the research object. Cell adhesive assay was used to determine the influence of quercetin on the HCCLM6 adhesive ability to extraeellular matrix. Immunofluorescence methods and Western blot was used to determine the expression of protein E-Cadherin in HCCLM6. Results Quercetin inhibited the adhesion of HCCLM6 to Marigel glue. In quercetin-treated group, the expression levels of protein E-Cadherin of HCCLM6 were higher than the control by immunofluorescence methods and Western blot. Conclusion Quercetin could inhibit the adhesive ability of high-metastatic hepatoma cells. The mechanism may be related to upregulating the expression of protein E-Cadherin in hepatoma cells.
