摘  要:目的:探讨α-磷酸三钙/胶原复合体修复大鼠头盖骨骨缺损能力。方法:选用8周龄SD大鼠28只,随机分为自体骨组(n=12)、α-磷酸三钙/胶原复合材料组(n=12)与空白组(n=4)。自体骨组和材料组于术后4、6、8周3个时间点各处死大鼠4只;空白组大鼠在8周全部处死。分别进行mirco-CT扫描分析,HE、TRAP和ALP染色,比较各组修复骨缺损效果。结果:8周时α-磷酸三钙/胶原复合物和自体骨的骨体积分数分别为(69.7±3.95)%和(42.475±4.38)%(P<0.05);随着时间延长,α-磷酸三钙/胶原复合材料逐渐被吸收,同时骨缺损亦逐渐修复;自体骨缺损部在6周后出现大量破骨细胞,大面积移植骨与新生骨被吸收,而α-磷酸三钙/胶原复合体修复骨缺损中较少发现破骨细胞。结论:α-磷酸三钙/胶原复合体具有较好骨缺损修复能力,且材料易于吸收。AIM:To study the effects ofα-tricalcium phosphate(α-TCP)/atelocollagen sponge composite in the repair of calvarial defects in rats.METHODS:Bone defects (diameter,8 mm;depth,1 .0 mm)were created in 28 male SD rats aged 8 weeks .The rats were divided into 3 groups.Inα-TCP group(n=12)the bone defects were filled with α-TCP /atelocollagen sponge composite,in positive control group (n=12)with autogenous bone,in blank control group (n=4)with nothing.The rats in α-TCP and positive control groups were euthanized at 4,6,and 8 weeks after surgery respectively(n=4).The rats in blank control group (n=4)were euthanized 8 weeks after surger-y.The calvarial bone was scanned by micro-CT.HE TRAP and ALP staining were performed for the evaluation of new bone formation.RESULTS:Micro-CT images at 8 weeks after surgery revealed that,in α-TCP and positive control groups the bone volume was as (69.7 ±3.95)%and (42.475 ±4.38)%respectively(P〈0.05).Histological anal-ysis revealed that in α-TCP group new bone contiguous with original bone was observed and the composite was ab-sorbed.In positive control group lot of osteoclasts and absorption of the transplanted autogenous bone were observed.In blank control group little new bone was observed at the boundary of the deffects.CONCLUSION:α-TCP /atelocolla-gen sponge composite is effective in the repair of bone defect.
