NF-κB活化在肠缺血再灌注全身多脏器的表达及意义

摘　　要：目的观察肠缺血再灌注后细胞核因子-κB（NF-κB）蛋白的表达及作用。方法 C57BL/6小鼠被随机分为假手术组（sham组,简称S组）和缺血再灌注组（II/R组）,观察肠缺血再灌注后肠、肝、肺和肾脏组织病理学改变,电泳迁移率变动分析法（EMSA）检测肠、肝、肺组织NF-κB活性表达。结果 （1）肠缺血后再灌注6h肝、肺和肾脏组织发生明显病理学改变;（2）II/R后肠、肝、肺组织NF-κB活化。与假手术组比较,肠组织NF-κB持续活化,尤以再灌注4h、6h升高明显（P〈0.01）;在肝脏组织,NF-κB活化表现呈升高—降低—再升高双峰型特点,再灌注0.5h、6~12h NF-κB活化明显（P〈0.01）;在肺组织,肠缺血期NF-κB即活化,再灌注1~4h及12hNF-κB活化明显（P〈0.01）,表现呈升高—降低—再升高重复活化特点。结论 NF-κB活化参与了肠缺血再灌注全身多脏器损害。Objective To investigate the expression of activated nuclear factor kappa B （NF‐κB） protein after in‐testinal ischemia‐reperfusion and study the effect of NF‐κB protein on intestinal ischemia/reperfusion （II/R） injury in mice .Method Intestinal ischemia/reperfusion was induced by occluding the superior mesenteric artery for 40 mins followed by reperfusion .C57 BL/6 mice were divided randomly into sham‐operated group （sham group） and II/R group .The intestine ,lung ,liver and kidney pathology was observed by optical microscope in 6h after reperfusion in II/R group or sham group .NF‐κB activation levels of nuclear protein were measured using EMSA analysis in the in‐testinal ,hepatic and renal tissue after II/R .Result After II/R induced NF‐κB activation of intestinal ,hepatic and pulmonary tissue .Compared with sham group ,intestinal NF‐κB activation induced by II/R significantly increased , especially in 4‐6 h after reperfusion （P〈0 .01） .Hepatic and pulmonary NF‐κB activity rapidly increased ,tempora‐rily reduced and obviously elevated in early period of intestinal ischemia or reperfusion .Therefore ,NF‐κB activation of the liver and lung showed characteristic of repeated activation in mice of II/R .Conclusion NF‐κB activation is in‐volved in systemic multiple organ damage induced by II/R in mice .
