摘  要:目的:研究连花参加方对4-硝基喹啉-1-氧化物(4-nitro quinoline-1-oxide,4NQO)诱导C57BL/6小鼠食管癌前病变形成过程中β-catenin蛋白表达的影响,并探讨其抑制食管癌前病变可能的作用机制。方法 :采用免疫组织化学法检测4NQO诱导小鼠食管癌前病变不同阶段以及给予连花参加方治疗前后各组小鼠食管组织中β-catenin蛋白表达水平的变化。结果 :β-catenin蛋白在单纯诱癌组小鼠食管鳞状上皮细胞中的异常表达率随着诱癌时间的延长而升高。诱癌开始后第24周时β-catenin蛋白的异常表达率为62.5%(10/16),显著高于同期正常对照组(P<0.01)。与单纯诱癌组相比,连花参加方治疗组和全反式维甲酸(all-trans retinoic acid,ATRA)治疗组小鼠食管上皮组织中β-catenin蛋白的异常表达率均有不同程度的降低,差异具有统计学意义(P<0.01),但连花参加方和ATRA治疗组之间的差异无统计学意义。结论 :连花参加方可以抑制β-catenin蛋白的聚积,这可能是其抑制小鼠食管癌变发生与发展的机制之一。Objective:To investigate the effect of Lianhua Shenjia Formula(this formula is extracted from the effective components of four traditional Chinese medicines:phillygenin,trichosanthin,codonopsis pilosulaglycoprotein and triterpenes compound of Cortex Periplocae) on p-catenin protein expression during the process of esophageal precancerous changes in C57BL/6 mice induced by 4-nitro quinoline-1-oxide(4NQ0),and to explore the possible mechanism of this formula inhibiting precancerous changes in esophagus.Methods:The expressions of p-catenin protein in different-stage esophageal precancerous lesion tissues in mice induced by 4NQ0 and before and after Lianhua Shenjia Formula treatment were detected by immunohistochemistry.Results:The abnormal expression rate of p-catenin protein in esophageal squamous cells was increased over time in 4NQ0 control group,and this rate reached 62.5%(10/16) in the 24 th week,which was significantly higher than that in the normal control group(P 0.01).The abnormal expression rates of p-catenin protein in Lianhua Shenjia Formula treatment group and all-trans retinoic acid(ATRA) treatment group were both decreased as compared with that of 4NQO control group(P 0.01);however,there was no significant difference between Lianhua Shenjia Formula treatment group and ATRA treatment group.Conclusion:The Lianhua Shenjia Formula can inhibit the accumulation of p-catenin protein,and this may be one of the mechanisms of Lianhua Shenjia Formula inhibiting the development and progression of esophageal carcinoma.
