摘  要:目的评价XRCC1 Codon399单核苷酸多态性与非小细胞肺癌患者晚期放射性损伤的相关性。方法选取经皮肺穿刺活检病理确诊为鳞癌、腺癌、腺鳞癌或大细胞癌患者60例,以x TAG液相芯片技术检测XRCC1 Codon399的3种基因型。随访至放疗结束后6个月,评价晚期放射性损伤与不同基因型的关系。结果 Arg/Gln杂合子组患者发生晚期放射性肺损伤的比例高于两组纯合子(P<0.05),Arg/Gln和Arg/Arg组患者出现心脏损伤的比例高于Gln/Gln纯合子组(P<0.05)。各组患者的皮肤、食管损伤的比例差异无统计学意义(P>0.05)。结论 XRCC1 Codon399单核苷酸多态性与晚期NSCLC患者肺部及心脏的晚期放射性损伤有关,有望成为放射治疗副作用的预测因子之一。Objective This study aimed to investigate the association between single nucleotide polymorphism of XRCC1 Codon399 and late radiation injury in normal tissues of patients with non-small cell lung cancer(NSCLC). Methods 60 patients with pathologically confirmed squamous carcinoma, adenocar- cinoma, adenosquamous carcinoma or large cell carcinoma were respectively analyzed. Get the genotype of XRCC 1 Codon399 with xTAG liquid chip, evaluate the late irradiation-induced injury in six months after radio- therapy and compare the late radiation reactions of normal tissue in the different genotypes. Their correlations were statistically analyzed with chi-square test(P 〈0. 05) was considered statistically significant. Results The patients with Arg/Gln heterozygote had a higher rate of late radiation-induced pulmonary injury than the others( P 〈 0. 05 ), meanwhile the patients with Arg/Gln heterozygote and Arg/Arg homozygote were easier to injury heart (P 〈 0. 05 ). There were no significant differences among the three genotypes involved late irradia- tion-induced injury with skin and esophagus (P 〉 0. 05 ). Conclusions The single nucleotide polymorphism of XRCC1 Codon399 is associated with late irradiation-induced injury wit pulmonary and heart in patients with advanced NSCLC. It is expected to be one of the predictive factors with the side effect of radiotherapy.
