摘  要:中度嗜盐菌胞外多糖对逆环境具有一定的耐受性,这种抗逆性的微生物絮凝剂在玉米淀粉加工废水处理中具有广阔的应用前景。本文从盐池底泥中筛选出具有较高絮凝活性的菌株;16S r DNA鉴定为Halomonas sp.H09;红外光谱分析表明该菌株产生的絮凝活性物质为多糖。Halomonas sp.H09多糖发酵的优化条件为培养基Na Cl浓度为30g/L,初始p H为7.0,60g/L的葡萄糖为碳源,15g/L的(NH4)2SO4为氮源。在优化的条件下Halomonas sp.H09多糖合成量为3.7g/L。Halomonas sp.H09多糖在玉米淀粉加工废水絮凝活性测定体系中,最适多糖添加量为0.015%,最适絮凝温度为30℃,20min絮凝率最高达到88.7%,该条件下絮凝处理后固形物,浊度、色度和CODcr的去除率分别达到85.8%、72.4%、77.8%和69.3%。Halomonas sp.H09多糖应用在玉米淀粉加工企业废水的处理中,具有添加量少、絮凝热稳定性较好等特点,具有较高的应用优势。The polysaccharide produced by moderately halophilic bacteria was relatively well tolerated to adverse environment, the resistance of the microbial flocculants in the flocculation treatment of wastewater in the corn starch processing has a broad application prospect. A bioflocculant-producing bacterium named Halomonas sp. H09 was isolated and screened from salt sludge. The polysaccharide fermentation conditions were optimized. Effect of flocculation conditions on flocculation activity of Halomonas sp. H09 polysaccharide was investigated. The optimal conditions for polysaccharide fermentation were 30g/L NaC1 and initial pH of 7.0. The optimized carbon source was 60g/L glucose and nitrogen source was 15g/L (NH4) 2SO4. The polysaccharide concentration as high as 3.7g/L was achieved under the optimal conditions. In the system of the corn starch processing wastewater flocculating activity determination, the highest flocculation rate was 88.7% on 20rain, with polysaccharide addition of 0.015% at 30 ~C . Under above conditions, the solids removal rate was 85.8%, the turbidity removal rate was 72.4%, the chroma removal rate was 77.8% and CODor removal rate was 69.3%. In the treatment of corn starch wastewater processing enterprises, Halomonas sp. H09 polysaccharide had the advantage of lower additive amount, higher flocculation thermal stability, and thus applied widely.
