摘 要:目的从能量代谢角度比较二神丸中补骨脂、肉豆蔻炮制前后对脾肾阳虚泄泻大鼠的调控效应。方法采用腺嘌呤灌胃,饮食失节,服冰番泻叶水浸剂方法复制脾肾阳虚泄泻大鼠模型,给药二神丸进行治疗性实验,酶标仪法检测大鼠肝组织中Na+-K+-ATPase,Ca2+-Mg2+-ATPase以及乳酸脱氢酶(LDH)活性变化;比色法检测肝组织中琥珀酸脱氢酶(SDH)活性变化。结果与正常对照组比较,模型组肝组织Na+-K+-ATPase、Ca2+-Mg2+-ATPase及SDH活性显著降低(P<0.01);LDH活性显著升高(P<0.01)。与模型组相比较,二神丸Ⅳ组(盐炙补骨脂+麸煨肉豆蔻)Na+-K+-ATPase、Ca2+-Mg2+-ATPase及SDH活性明显升高,LDH活性明显降低,均具有极显著性差异(P<0.01);与二神丸Ⅰ组(补骨脂生品+肉豆蔻生品)相比较,二神丸Ⅳ组Na+-K+-ATPase、Ca2+-Mg2+-ATPase及SDH活性均高于二神丸Ⅰ组,差异有统计学意义(P<0.05);LDH低于二神丸Ⅰ组,差异有统计学意义(P<0.05)。结论经盐炙补骨脂和麸煨肉豆蔻组方的二神丸改善脾肾阳虚泄泻模型大鼠能量代谢效果更加明显,提示其增效可能与促进能量代谢相关酶活性有关。AIM To compare the effects of after and before processing of Buguzhi (Psoraleae Fructus) and Roudoukou (Myristicae Semen) which comprised Ershen Pills on rats with spleen-kidney yang deficiency diarrhea from the viewpoint of energetic metabolism. METHODS The rats models for the spleen-kidney yang deficiency diarrhea were made through the perfusion of adenine, irregular diet, and reception of ice senna extract. The models were treated with Ershen Pills. The activity of Na^+ - K^+ -ATPase , Ca^2+ -Mg^2+ -ATPase and lactate dehydrogenase (LDH) in the liver tissue were measured by enzyme-linked immunosorbant assay. And the activity of succinate de- hydrogenase (SDH) was observed by the colorimetric method. RESULTS In comparison to the normal control group, processed Buguzhi and Roudoukou expressed a lower level of Na ^+ -K^+ -ATPase ( P 〈 0. 01 ), Ca^2+ -Mg^2+ ATPase (P 〈0. 01) and SDH (P 〈0. 01), and a higher level of LDH (P 〈0. 01 ) in model group. Compared with the model group, the Ershen Pills 1V group could enhance the level of Na^+ -K^+ -ATPase, Ca^2+ -Mg^2+ -ATPase and SDH activity, decrease the level of LDH activity. There were extremely significant differences (P 〈 0. 01 ). The Na^+ -K^+ -ATPase, Ca^2+ -Mg^2+ -ATPase and SDH activity in Ershen Pills IV group were higher than those in Ershen Pills I group, the difference was statistically significant (P 〈 0. 05 ). The LDH activity in Ershen Pills IV group was lower than that in Ershen Pills I group, the difference was also statistically significant ( P 〈 0. 05 ). CONCLUSION Ershen Pills, consisting of salt-frying Buguzhi and bran-roasting Roudoukou, has the function of improving the energetic metabolism of rats with spleen-kidney yang deficiency diarrhea, which may be related to relevant enzymes.
